| Objective:This thesis aims at finding the impacts and mechanism of different dosages of salt in food, illustrating the meddling effect of Danshen in promoting blood circulation and removing blood stasis, and providing more supporting evidences with traditional Chinese medical application and Chinese theory--limited-salt and caring for life.Methods:Theory-based and empirical methods are applied in this study.1. Theory-based study:Based on the reviews, such as the concepts of salt, blood circulation, blood stasis in traditional Chinese medicine and the theory "Salt restrains blood", this paper analyzes the relationship between salt and blood, internal organs and medicine to find the mechanism of this theory in traditional Chinese medicine and its application in clinical practice.2. Empirical study:First, all of the samples-healthy SD mice, are divided into six groups:No-salt Group (NG); Low Dosage Salt Group(LG); Intermediate Dosage Salt Group I(IGI); Intermediate Dosage Salt Group Ⅱ(IGII); High Dosage Salt Group(HG); and Danshen Group(DG). Second, all of the samples are given a enema with12%NaCl except the NG for30days:for LG,0.5mLNaCl per100g every day; for IG,1mLNaCl per100g every day; for HG,2mLNaCl per100g every day; and for DG,1mLNaCl per100g every day. Third, for DG, it is injected with Danshen liquid seven days before the molding is over; for IGII, it is fed with distilled water for15days from the31st day. Through the whole process, the quantity of water drinking, weight, and the colors of their lips, tongue and pawls are detected. When this test is over, it focuses on the changes of veins under their tongues, and six mice in each group are arbitrarily chosen to see their blood flowing deformation after they are killed. Finally, ELISA is used to detect the activity of their ET, NO, IL-6and TNF-a. These dead animals’coronary arteries are taken out to collect mRNA on an ultra-clean table and back-transcript into cDNA. Using the qRT-PCR, it is to find how the thrombus conditions mRNA (TMmRNA) level and see how the pathology and shape of coronary arteries change under the light microscope.Results:1. The results from literature review:1) Salt and blood, organs and the herbs have an intricate relationship.2)The theory "Salt restrains blood" in Neijing shows that salt may be harmful to blood. TCM mechanism for the occurrence of salt or salty cold, diarrhea, blood stagnation; salts and damage of kidney, liver and lungs.2. The results from empirical study:1) The symptom that tongues and pawls turn purple are first discovered in HG;2) In IG, especially in HG, collaterals which connects branches of channels under tongue are extended, darker and more branches can also be found;3) Compared with that in the NG, blood stickiness, ET, NO, IL-6and TNF-a in IG and HG are significantly different(P<0.01, P<0.05), but in LG, no significant difference can be found (P>0.05), and Danshen can reduce the blood stickiness and the levels of ET, NO, IL-6and TNF-a (P>0.05);4) Compared with that in the NG, TMmRNA in coronary artery is high (P<0.01) in IG, but in LG, no significant difference can be found (P>0.05), and the TMmRNA in DG is lower than that in NG, but not significant (P>0.05);5) Acute inflammation of coronary artery can be easily found in IG and HG, and the phenomenon that artery vessels become thicker also can be found in IG, while in LG and DG, this symptom is not obvious except artery vessels become a little thicker.Conclusion:Salt influences the function of blood, and salt can be one of the factors promoting blood circulation. Intermediate and high dosages of salt are more harmful to blood compared with the low dosage, and Danshen can improve the conditions of blood. It also proves that it is feasible to establish the prototype of curing blood stasis by using12%NaCl. This paper provides evidence of the theory "Salt restrains blood" in Neijing, supports the Chinese theory---limited-salt and caring for life, and also gives new ideas and ways for Chinese way of treating blood stasis. |
