| Objectives:To investigate the curative effect, quality of life, adverse reactions on curing esophageal cancer combined radiotherapy and explore the mechanism of the action on inflammatory microenvironment of esophageal cancer and angiogenesis by liushen pill.Method:(1) The clinic study:30patients with esophageal cancer were divided into experimental group and control group. The control group was only treated by radiotherapy, and the experimental group was treated by liushen pill combined radiotherapy. After a period of treatment, short-term curative effect and the response rate and toxicity and QOL were compared in two groups.(2) The experimental study:ESCC xenografts in nude mice was established by injecting ESCC cell suspension subcutaneously, and then intervented by liushen pill, dynamic observed the tumor growth in different groups, record living conditions, measured the tumor volume, draw growth curve; And the tumor-suppressing rate was calculated; Immunohistochemistry was used to detect microvessel density (MVD), and determination of TNF-α, IL-6, NF-κB, VEGF level by RT-PCR and Western-blot.Results:(1) The clinic study:There is no significant differences in the recent curative effect, but compared the control group, the experimental group improved the quality of life more obviously, and significantly lower in the incidence of adverse reactions.(2) The experimental study:Liushen Pill can inhibit the growth of the human Esophagus squamous cell carcinoma xenografts. The tumor-suppressing rate in liushen pill groups was much higher than that of the model group(P<0.05); Compared with model group, the difference between model and liushen pill groups was significant (P<0.05). The expression of TNF-α, IL-6, NF-κB was lower in group of liushen pill compared with model group; The microvessel density (MVD) in the treatment group was much lower than that of the model group.Conclusion:(1) The clinic study:That cannot improve the short-term curative effect on the patients with esophageal cancer by liushen pill combined radiotherapy, but compared with radiotherapy alone, it can significantly improve the patients quality of life, reduce the occurrence of adverse reactions.(2) The experimental study:Liushen Pill can inhibit the growth of the human esophagus squamous cell carcinoma xenografts. The treatment with Liushen Pill can effectively down-regulate the expression of VEGF and MVD, then inhibit the angiogenesis, which is one of the tumor-inhibiting mechanism of Liushen Pill. Liushen Pill can inhibit tumor by bringing down the expression of TNF-α, IL-6, NF-κB and the TNF-α-NF-κB signaling pathway in inflammatory microenvironment of esophageal cancer which may be the one of the antitumor mechanism of Liushen Pill. One of the tumor-inhibiting mechanism of Liushen Pill is inhibition of tumor angiogenesis by regulating the TNF-α-NF-κB signaling pathway. |
