The Research Of The Mechanism Of Bushen Jianpi Huoxue Decoction In The Diabetic Osteoporosis Rats Based On The NF-kappa B And Wnt Signaling Pathways

Objective: To explore the mechanism of Bushen Jianpi Huoxue Decoction treating diabetic osteoporosis at the molecular level by observing the changes of the related blood biochemical indexes of bone metabolism 、bone mineral density(BMD)、the correlation factors’ expression of NF-kappa B and Wnt signaling pathways in bone tissue.Methods: 55 male Wistar rats were used as the model of the study,10 rats were divided into normal control group,other 45 rats were chosen to establish the models of diabetic osteoporosis by using the combination of high-carbohydrate-high-fat diet and intraperitoneal injection of streptozotocin. After measuring blood glucose and bone mineral density, the rats which conformed to the diagnostic standard of DOP were randomly divided into Chinese traditional medicine group,the western medicine control group and model group, Bushen Jianpi Huoxue decoction, Calcium Carbonate+AlPhaD3 and physiological saline were administered respectively by gavage. All rats were sacrificed at the end of 12 weeks to detect the levels of fasting blood glucose, insulin, serum calcium, phosphorus, alkaline phosphatase and the BMD of femoral in each group,at the same time, to detect the expressions of Runx2, LRP5, β-catenin, TRAF6, NF-kappa B, NFATc1 mRNA in the rats’ bone by using the method of reverse transcription-polymerase chain reaction(RT-PCR).Results: The levels of fasting blood glucose and insulin in post-treatment were all lower than those in pre-treatment in the Chinese traditional medicine group. There were statistically significant in the level of fasting blood glucose and insulin(P<0.05)before and after the treatment. Compared with the model group, the levels of fasting blood glucose 、insulin、 phosphorus、 alkaline phosphatase in the Chinese traditional medicine group were all decreased and the bone mineral density was increased after treatment, those all had high statistical significance(p< 0.01).After treatment,the fasting blood glucose and bone density in the Chinese traditional medicine group all had statistical significance difference(p < 0.05) compared with the western medicine control group, and the insulin had highly significant difference(p < 0.01). After treatment, the expressions of Runx2, LRP5, β-catenin mRNA in the rats’ bone were all increased in the Chinese traditional medicine group, which all had high statistical significance difference compared with the model group(p<0.01), and statistical significance difference compared with the western medicine control group(p < 0.05). After treatment, the expressions of TRAF6、NF-κB、NFATc1 m RNA in the rats’ bone were all decreased in the Chinese traditional medicine group, which also had high statistical significance difference compared with the model group(p<0.01).There were statistical significance difference(p<0.05) in the expression of TRAF6 mRNA and high statistical significance difference(p< 0.01)in the expressions of NF-κB、NFATc1 mRNA between the Chinese traditional medicine group and the western medicine control group.Conclusion: Bushen Jianpi Huoxue Decoction could reduce the level of fasting blood glucose, improve insulin resistance and increase bone density, its curative effect is superior to the western medicine control group. Wnt and the NF-kappa B signaling pathway could influence the type 2 diabetic osteoporosis rats’ bone reconstruction, which were important molecular biological mechanisms of this disease. Bushen Jianpi Huoxue Decoction could treat the diabetic osteoporosis by activating the Wnt signaling pathways and inhibiting the NF-kappa B signaling pathway.
