| Objective:To observe efficacy of ShiReQing(SRQ) for perianal eczema and impact on expression of the mRNA of transcription factor T-bet/GATA-3.Explore the possible therapy mechanism associated with Th1/Th2 cells of SRQ in perianal eczema.Method: 40 patients from Shandong Provincial Hospital of Traditional Chinese Medicine and Affiliated Hospital of Shandong University of Traditional Chinese Dermatology, were randomly divided into two groups---group with SRQ and group with KuShenPian. Both of the group were given Xiaofansan for external use on perianal eczema. 4 weeks were a course of treatment. Respectively, before and after treatment the scores of the area, pruritus etc were recorded and analyzed. The blood of patient were detected to study the expression levels of T-bet/GATA-3 mRNA.Establish a mouse chronic eczema model with different concentrations of DNCB. investigate skin lesions and pathological changes to find the optimum concentration of DNCB and study the curative effect of Shireqing for chronic perianal eczema. There were six groups: normal control group, model group, Shireqing low, medium and high dose groups and Longdanxiegantang groups as the positive control. After the fourth 24 h of excitation, skin reactions were observed and scored according to the severity of erythema. Low, medium and high concentrations of decoction of Shireqing were prepared and daily gavage was 0.1ml/10 g. One week after the treatment morphological changes, symptoms were scored the last of the two set of data for statistical analyzed.Result: Shireqinghad significant effect forperianal eczema compared with control group(P <0.05). The result of histopathological staining illustrated that IL-2, IL-4 was significantly decreased, while IFN-γ was increased in experimental group. PCR andWestern showed after the treatment of Shireqing the expression of T-bet decreased obviously and T-bet/GATA-3 proportion tends to be normal.(P< 0.05).Conclusion: Shireqing could be a effective therapeutic for perianal eczema. It significantly changed T-bet/GATA-3 proportions to restore the balance of Th1/Th2 cell by interfering with upstream transcription factor, which can be used as therapeutic for perianal eczema and T-bet/GATA-3 could be an ideal target molecule. |
