The paper mainly researched the velamen of Dictamnus dasycarpus Turcz and the fruit of Siraitia grosvenorii. Through modern extraction, separation and purification method, this paper also researched the main components in the study and the effect of dictamnus on anti-inflammatory and the effect of Siraitia grosvenorii on acute pneumonia.According to different extraction methods and single factor experiment, this paper extracted the oil with supercritical extraction technology, and designed Box-Behnken experiment with Design-Expert software. The result showed the productive rate of oil and fat from the cortex dictamni was 1.2942%, and 4.296% of Momordica grosvenori. Compared with the prediction of experimental design, this method illustrated that the fitting degree of the model was high and could accurately reflect the extraction pressure, time and temperature on the cortex dictamni; FT-IR and GC-MS analysis showed 74 spectral peaks of cortex dictamni and 61 spectral peaks of Momordica grosvenori. According the literature, the detection component library spectra and NIST05 a.L standard for comparison, this paper identified respectively 20 and 25 components.Based on the extraction method and single factor test, this research took dictamnine and Mogroside V as target components through ASE technology. According to optimized the experimental conditions by response surface method, the result showed that the average content of Dictamnine and Mogroside V was respectively 14.16% and 10.02%; This result was close to predictive value, and it reflected the effect of influence factors on extraction efficiency of Dictamnine and Mogroside V. Using fast solvent extraction(ASE) and ultra high performance liquid chromatography and three level four tandem mass spectrometer(UPLC-TQMS) technology analyzed their main chemical component, and establish the identifying and analytical method for Dictamnine and Mogroside V. The experimental results showe dictamnine, fraxinellone, obacunone and bitter lemon pigment content were respectively 151.25mg/g, 213.25mg/g, 121.17mg/g and 687.5mg/g, Mogroside V, Momordica grosvenori saponin IV, Momordica grosvenori saponin III and Mangosteen 11-O- saponin V content wer respectively 121.25mg/g, 42.25mg/g, 49mg/g and 623.5mg/g. Experimental results showed the highest content of saponins in Momordica grosvenori 11-O- Mogroside V, secondly for Mogroside V and two other soap glucoside content relative were low.With accelerated solvent extraction(ASE) and high speed countercurrent chromatography(HSCCC), this paper analyzed the dictamni main components. According to the results of HSCCC parameter optimization, the final separation parameters: HEX- EtOAc-MeOH-Wat er-BuOH(V:V:V:V:V, 0.8:1:1:1: 0.16) as a two-phase system, which is stationary phase, in order to mobile phase, the mobile phase. Applying the method from the medicinal plants of cortex dictamni separation purification of 4 kinds of compounds, and compounds were compared with HPLC standard alignment and HPLC-MS identification and analysis; and the main components was indentified as ultimately determine dictamnine(purity of 98.6%), fraxinellone(97.2% purity), obacunone(99.0% purity) and bitter lemon prime(98.4% purity).The research analyzed the anti-inflammatory activity of cortex dictamni through mice capillary permeability model, ear swelling model and granulation hyperplasia model. The results showed that cortex dictamnie and fraxinellone dose groups could significantly inhibit ear swelling caused by xylene, and the inhibition rate was high and the difference was significant(P < 0.05); medium and high dose of cortex dictamnie had clear effect on mice capillary permeability model;. The low and medium dose of fraxinellone had obvious effect on mice capillary permeability modelThis research separated and purified main components of Momordica grosvenori with RP-HPLC and analyzed the activity of Momordica grosvenori V on acute lung injury model. The anti-inflammatory mechanism was discussed.According to RP-HPLC method for preparation of two components by standard alignment and HPLC-MS analysis, this result wa identified as 11-O- Mogroside V(92%) and Mogroside V(90%).Lung injury model by LPS was successfully established, This paper observed effects of Mogroside V on the mouse model of lung injury effect; through HE staining, changes of levels of cytokines and lung wet dry ratio, MPO activity, this research showed that Mogroside V has inhibitory effect on acute lung injury. The research found mogrosides V had a certain effect on NF KB signaling pathway; iNOS and COX-2 expression were reduced, and the expression of T-IkBa was increased. Mogroside V inhibition effect on the lung injury in mice was finished through the inhibition of NF-KB signaling pathway.
