Effect Of Electroacupuncture With “Combined Biao And Ben Acupoints”on Myocardial Ischemia And Mechanism Of Mitochondria Signaling Pathway

ObjectiveUsed isoprenaline hydrochloride(ISO) to make model of myocardial ischemia in male rats of wistar.Our team had made lots of researches on myocardial ischemia with“neiguan”(P6) acupiont for over a decade.We found that this acupoint could against ha-rm of myocardial ischemia through many pathways.On base of these researches and according to traditional chinese medicine, t-his experiment chose electroacupuncture(EA1) with double“neiguan”(P6), double“zusanli”(ST36)and “guangyuan”(CV4)to contrast electroacupuncture with double“neiguan”(P6), double“shenmen”(H7),“danzhong”(CV17) acupoints in respect of treating myocardial ischemia, and to explore possible mechanism of EA1, which provided evidence for clinic treatment and choosing acupoints.MethodsA total of 50 SPF Wistar rats were randomly divided to 5groups,each having 10.Normal group: each rat was injected 0.9% Na Cl solution with 5mg/kg/d into back skin on each morning for 7 days,once a day. Model group: each rat was injected ISO solution with5mg/kg/d into back skin on each morning for 7 days, once a day, andtested ECG after experiment. If the height of S-T segment in ECG was higher than 0.1m V, the model of myocardial ischemia meat success.Before made model,both EA1 group and EA2 group were respectively taken double“neiguan”(P6), double“zusanli”(ST36), “guangyuan”(CV4)acupoints and double “ shenmen ”(H7), “ danzhong ”(CV17)acupoints connected with HANS-200 electric acupuncture instrument for 10 min, 2-100 Hz, density waves and 1m A on each morning, once a day, and this treatment lasted 14 days after made model; Before made model, resveratrol group was filled stomach with resveratrol solution with 5mg/kg/d on each morning, once a day, and this treatment lasted 14 days after made model. In the process of experiment, observed daily act of each group. The end, tested S-T and HR of ECG, c Tn I of blood, MDA, SOD, GSH-Px of heart tissue, and detected the protein and m RNA of SIRT1, PGC-1α, NRF-1 and mt TFA in heart tissue, and discriminated the supermicro pathological structure changes of heart cells. Analyzed these datas by statistical analysis system.Results1.Observation of daily act of each group: Normal group was all fine.In the process of experiment, rats of model group had heartbeat,shortness of breath, drowsiness when they were injected ISO solution in first day, and then emerged epilation, yellow and dry hair, white tail, purple mouth, depression, slowing to respond,loss of appetite when they were injected ISO solution in 7th day;Compared with rats of model group, rats among EA1 group, EA2 group and resveratrol group were all better, only a little adverse symptoms.2.Effect on S-T and HR of ECG: S-T segment of model group was significantly higher than that of normal group(P<0.01); Compared with model group, the height of S-T segment among EA1 group,EA2 group and resveratrol group were all significantly lower(P<0.05);No significant difference of S-T segment among EA1 group, EA2 group and resveratrol group(P>0.05). HR of model group was significantly faster than that of normal group(P<0.01); Compared with model group,HR among EA1 group,EA2 group and resveratrol group were all slower(P<0.05); HR of two EA groups were slower than that of resveratrol group(P<0.05).3.Effect on c Tn T of myocardial enzyme: The level of c Tn T in model group was significantly higher than that of normal group(P<0.01); The level of c Tn T among EA1 group,EA2 group and resveratrol group were all significantly lower than that of model group(P<0.05); The level of c Tn T in EA1 group was lower than that of EA2 group(P<0.05), but no significant difference between EA1 group and resveratrol group(P>0.05).4.Effect on supermicro pathological structure in heart cells:normal group,scardiac muscle fibers were arranged in neat rows, and mitochondria structure was intact; model group,s cardiac muscle fiber was disorder, muscle plasma nets expansion, parts of swelling mitochondria, shortening or disappearing crest; Compared with model group, the degree of harm among EA1 group,EA2 group and resveratrol were all lighter, which the number of mitochondria had grown, and the structure of mitochondria were better; The degree of harm in EA1 group was lighter than EA2 group.5.Effect on MDA, SOD, GSH-Px in heart tissue: Compared with normal group, the level of MDA in model group was significantly allhigher(P<0.01), but the level of SOD and GSH-Px in model group were significantly all lower(P<0.01). Compared with model group, the level of MDA among EA1 group,EA2 group, and resveratrol group were all lower(P<0.05),but the level of SOD and GSH-Px among EA1 group,EA2 group, and resveratrol group were all higher(P<0.05);Compared between EA1 and EA2, the level of MDA in EA1 group was lower than that of EA2 group(P<0.05), but the level of SOD and GSH-Px in EA1 group was all higher than that of EA2 group(P<0.05); No significant difference between resveratrol group and EA1 group in MDA(P>0.05), and no significant difference between resveratrol group and EA2 group in SOD and GSH-Px(P>0.05).6.Effect on the level of PGC-1α, NRF-1, mt TFA in heart tissue:Compared with the normal group,the level of PGC-1α, NRF-1 and mt TFA in model group were significantly all lower(P<0.01); Compared with model group, the level of PGC-1α, NRF-1, mt TFA among EA1 group,EA2 group, and resveratrol group were all higher(P<0.05); Compared between EA1 and EA2, the level of NRF-1 and mt TFA in EA1 group were all higher than that of EA2 group(P<0.05), but no significant difference between EA1 group and EA2 group in PGC-1α(P>0.05);The level of PGC-1αin resveratrol group was higher than that of two EA groups(P<0.05), and the level of NRF-1 and mt TFA were higher than that of EA2 group(P<0.05), but no significant difference between EA1 group and resveratrol group(P>0.05).7.Effect on the level of SIRT1 in heart tissue: Compared with the normal group, the level of SIRT1 in model group was significantly lower(P<0.01); Compared with model group, the level of SIRT1 among EA1 group,EA2 group, and resveratrol group were all higher(P<0.05); The level of SIRT1 in EA1 group was all higher thanthat of EA2 group(P<0.05), but no significant difference between EA1 group and resveratrol group(P>0.05).Conclusion1.EA could treat myocardial ischemia, but the effect of EA1 was better than that of EA2.2.EA1 could resisted the harm of myocardial ischemia through two pathway: activated SOD and GSH-Px, and reduced oxyradical to improve the ability of anti-oxidative damage in heart cells;regulated the signal pathway of SIRT1/PGC-1α/NRF-1/mt TFA/mt DNA.
