Association Studies Of Coagulation And Drug Resistance SNPs In The Avascular Necrosis Of The Femoral Head

BackgroundNowdays, the incidence of nontraumatic osteonecrosis of femoral head (ONFH) wass elevated, and the patients were younger than before. The ratio of hormone induced ONFH was also elevated. However, the pathogeny was not well known. Basing pathology research, the microthrombus was found in different ONFH patients.With the development of molecular biology, doctors foud the gene polymorphisy of coagulation and drug resistance had relationship with ONFH.Objects1. To identify the patients ratio of different ONFH types.2. To identify the relationship between candidate genes in the coagulation and drug resistance and ONFH with SNPs.3. To identify the relationship between the SNPs and different types of ONFH patients.4. To identify the relationship between SNPs and different races.MethodsAccording to the studies of ONFH, we chose the genes of coagulation (MTIIFR, F2, F5, N03and VEGFA) and drug resistance (ABCB1) as candidate genes. We genotyped35reported SNPs present in these genes among96ONFH patients (33hormone induced,29alcohol induced and34idopathic patients) and100healty controls with detailed clinical information. All the data of35SNPs with polymorphism were analyzed by the association analysis based on a single SNP, the association analysis between phenotypes and SNPs, and the association analysis between SNPs and differents ONFH types.Results1. There were no relationship between rs6541003, rs17037396, rs3766103, rs9332655, rs12131397, rs9332628, rs3766117, rs6427202、rs1894702, rs3766120, rs3136435, rs5897, rs3136526, rs3136516, rs3918156, rs35795287, rs3918185, rs743507, rs867225, rs25648, rs833069, rs3025033, rs1002204, rsl7149699, rs4148745, rs2235067, rs4148737, rs2235046, rs2235035, rs2235033, rs1202179, rs4148733, rs3213619and rs6465118and ONFH.2. There were no gene polymorphism in rs3918156, rs35795287, rs3918185, rs867225位点, rs5897and rs3136526.3. In the association analysis between SNPs and ONFH based on alleles, we got the postive SNP:rs2188526(ABCB1gene).4. In the association analysis between the genotypes and SNPs, I didn’t get any postive SNPs.5. In the association analysis between the SNPs and different ONFH types, I didn’t get any postive SNPs.Conclusion1. There were relationships between gene polymorphism and races.2. The rs2188526in the chromosome7were less in ONFH patients than healthy cortrols. 3. The genotypes of candidate SNPs were the same in the patients and controls.4. The candidate SNPs were the same in the different ONFH types.
