| Objective: Research the effect of "Qi lian fu zheng capsule" affecting proliferationand invasion of lung cancer A549cells by regulating of TGF-β pathway, and the clinicaleffect of "Qi lian fu zheng capsule" in maintenance treatment of advanced non-small celllung cancer.Methods: External cell culture experiments observe the effect of "Qi lian fu zhengcapsule"affecting the growth and proliferation of lung cancer A549cells from differentperspectives by using MTT, cell counting and HE staining. To observe the effect onmigration and invasion of lung cancer A549cells by scratch test and transwell experiment.To observe the expression of the factors of TGF-β pathway, such as TβRI, TβRâ…¡, Smad2/3, Smad4, Smad7, TGF-β1mRNA, E-cadherin and N-cadherin by using western blotting,RT-PCR and immunohistochemistry. Clinical research randomly divided the advancednon-small cell lung cancer patients into the observation group and the control group, theformer using "Qi lian fu zheng capsule" only, the latter giving the best supportive care, andcompared the tumor control rates, quality of life, PFS, OS,1-year,2-year survival, serumlevel of TGF-β1and drug safety of the two groups by follow-up.Results:"Qi lian fu zheng capsule" can inhibit the growth, proliferation, migrationand invasion of lung cancer A549cells, the effect expressed obvious at the high concentra-tion and combination with chemotherapy;"Qi lian fu zheng capsule" can raise the expre-ssion of TβRâ… , TβRâ…¡, Smad2/3, Smad4and E-cad, low down the expression of Smad7, TGF-β1mRNA and N-cad, correct disorders of TGF-β/smad pathway and prevent EMToccurrence."Qi lian fu zheng capsule" maintenance treatment of advanced non-small celllung cancer can improve the quality of the patients’ lives, prolong PFS (5.4month VS4.1 month), improve1-year survival rate and reduce the serum TGF-β1levels; but there is nosignificant advantage in improving tumor control rates, prolonging OS (12.3month VS10.6month) and improving2-year survival rate.Conclusion:"Qi lian fu zheng capsule" can prevent the invasion and metastasis oflung cancer A549cells by regulating the TGF-β pathway. It will be a feasible theapplication to maintenance treating the advanced non-small cell lung cancer. |
