6 male sterile lines of alfalfa were taken as the female parents,14 breeds of alfalfa with excellent properties as the male parents,and 84 hybridized combination were made using incomplete diallel cross.Then higher heterotic combinations were selected through the related analysis on the yield combining ability,cross-heritability and heterosis.High-producing and heterotic of alfalfa heterotic group which can be used classify in the way of SSR molecular notation and with the indicator of genetic distance.Further,in the 3 stages of pollen development,transcriptome sequencing,gene function annotation and expression pattern analysis were performed of male fertile and sterile flower buds.The results were as follows:(1)No.2 male sterile line and male parents II,III showed higher general combination ability,and the farther genetic distance between male and female parents could cross-fertilize higher heterotic combination.(2)Among 84 hybridized combination,heterotic effect and hybrid heritability showed in higher heterotic combinations,and the relation between the genetic distance and heterotic effect showed significant positive correlation,and when the cross parents whose genetic distances were in the range 0.3636 to 0.8182 notated by SSR were matched,the best performance was shown in heterosis,and the farther the genetic distances,the higher the specific combining ability of hybridized combinations.(3)The relation between the hybrid yield performance and water use efficiency showed significant positive correlation(P<0.05),and hybridized combinations 2×II,2×III,10×M6 and 13×M2 had higher content of crude protein in the analysis of nutrient quality character.(4)Through the homologous analysis on the Male fertile and sterile flower buds transcription group,Among them,73.76 % sequences had higher homology with Medicago truncatula.By gene function annotation and expression pattern analysis,it was confirmed that significant difference exists in the plant hormone signal,circadian rhythm,carbohydrate metabolism,flavonoids biosynthesis,pollen development,transcription factor and etc.
