Objective:To investigate the correlation of the content variation of cytokines(IL-8,18,10)from serum samples of patients with different levels of mycoplasma pneumoniae pneumonia(MPP)and the corresponding clinical inflammatory outcomes(CRP,LDH),which approachs the role of inflammation in the pathogenic mechanism of MPP,especially SMPP.Methods: The total of sixty-six cases from outpatients and hospitalized cases of the First Affiliated Hospital of Anhui Medical University between March 2013 and February 2015 were recruited,with thirty-four cases of MPP as the experimental group,and thirty-two age-matched and healthy cases as the control group,The cases of the experimental group were further divided into three groups: MPP cases with mild symptoms without concurrent asthma(Group 1);MPP cases with more severe clinical symptoms and without concurrent asthma(Group 2);MPP cases with mild symptom and asthma(Group 3).At the same time,clinical data were collected including the following information: gender,age,LDH,WBC,CRP,lymphocytes,neutrophils.The research aims to investigate the difference of cytokines and the inflammation outcomes between the two groups.The level cytokines were detected by ELISA method.Results:(1)Compared with the control group,The level of cytokines of IL-8,IL 10,IL-18 in the experimental group rised,the difference was statistically significant(P values were 0.044,< 0.001,< 0.001).(2)Compared with before treatment,the cytokines of IL-18 and CRP decreased in experimental group,the difference was statistically significant(P values were 0.020 and 0.001 respectively).(3)Compared with Group 1,IL-8 in the Group 2 had a higher level,which was not statistically significant(P = 0.232 >0.05).Compared with Group 1,The level of IL-10 in Group 2 decreased,and the difference was statistically significant(P < 0.05).Compared with Group 1,The level of IL-18 in Group 2 had a higher level,which was also statistically significant(P < 0.05).(4)The correlation analysis of the cytokines and clinical data showed that only IL-8 and IL-18 had correlation with CRP,with correlation coefficient of 0.467 and 0.619,respectively(P<0.001,P<0.01).The correlation coefficient was 0.223 between IL-10 and CRP,which was not statistically significant.Conclusion: The cytokines of IL-8,IL-18 and IL-10 take part in the inflammatory response of MPP,IL-10 plays a two-way adjustment role in the pathogenesis of the MPP.IL-10 can be used as one of inflammation predictor of MPP.Therefore,it is very important to control inflammation in the MPP,especially in SMPP.Objective: This research prepared to use animal experiment to demonstrate the effect of TNF-? and IL-8 on agonist-induced mouse trachea contraction,to verify the role of cytokines in the pathogenesis of MPP furtherly.Methods: We used organ culture to incubate mice trachea of 18 hours with TNF-? or IL-8.Then we detected the changes of agonist-induced tracheal smooth muscle contraction.The purpose of the study is to reveal the effect of TNF? and IL-8 on VDCC-mediated Ca2+influx in mouse tracheal smooth muscle,moreover,to investigate the change of VDCC protein and its effect on the tension of trachea.Results: In the present study,we found that:(1)TNF-? and IL-8 pretreatment can significantly enhance mouse tracheal contraction induced by KCl and carbachol;(2)VDCC channel blockers could significantly reduce the enhanced mice tracheal contraction pretreated by TNF-? and IL-8;(3)In Western blot,TNF-? and IL-8pretreatment significantly enhanced the expression of VDCC in mice trachea.Conclusion: These results suggest that inflammatory cytokines can enhance tracheal smooth muscle response to contractile factors by increasing the expression of VDCC,eventually which may cause bronchospasm and respiratory dysfunction.The study is helpful to understand the possible role and mechanism of inflammatory cytokines in the pathogenesis of MPP,and to provide a theoretical basis for future treatment,targets for drug screening and a new potential therapeutic approach for clinical treatment.
