POP is one of pelvic floor dysfunction(Pelvic Floor Dys function,PFD)dis-eases in older women,and it affect the quality of life very much.which is mainly composed of pelvic support defects or degeneration,injury and dysfunction caused by.The rapid development and application of high-throughput sequencing technologies,provides a more comprehensive and rapid analysis method for the study ofdisease pathogenesis,the research at present by gene chip technology for POP women with uterine cardinal ligament tissue(Uterosacral ligaments,USLs)gene expression differences were studied by RNA-Seq method,but theresearch on the POP women’s USL transcription groups of data has not been reported.Objective: To explore the pathogenesis on pelvic organ prolapse and keyregulation of mi RNA and its regulation of target genes to explore for research purposes,using high-throughput transcriptome sequencing tech nology,traditi onal me thod using classical experimental validation,screening of genes associated with the occurrence of POP,and in POP gene differential expression,and to explore the mechanism of pathway,and search for the tofunctionally important genes from the molecular level,in order to more fullyinvestigate the pathogenesis of POP pathology,to provide a scientific basisfor the treatment of relevant molecular targets for diagnosis,further.Methods: Gene expression between between the RNA-Seq technology POP compared with control and different period differences,and functional analysis of gene expression of these differences by using KEGG,GO,Bio GRID and other biological information database,to obtain the effect of key genes in the occurrence and development of POP and the signaling pathways important differences s in different periods,the other based on POP gene expression,screening that play a key role in the different periods of gene.In 5-8 patients with uterine ligament tissues of patients with pelvic organ prolapse of the sample,as well as in patients with peripheral blood samples for analysis of high flux transcriptome sequencing,usi ng biological informationinternationally recognized the combination of network bi ology theory to thedata comprehensively,the system of data mining and data integrationmethod,at the same time,with the classic method of traditi onal experimental verification changes in the expression of genes encoding,reveal the whole transcriptome level.Results:(1)The extraction of total RNA by quantitative and electrophoresis detection,OD260/OD280 is 1.8-2.2,reach RNA-seq sequencing sample require ement s.(2)By gene enrichment analysis,in the pelvic cavity organ,De,vertical patients,peripheral blood,gene enrichment,power,energy,mainly concentrated in the nerve signal transfer,delivery pathway and the TGF-beta signaling pathway,ce ll by cell,sub-fine,because,sub receptor,interaction,swallow,bite body,and cell ad hesion molecules,pathways.(3)from the normal to the pelvic organ prolapse of II degrees,II degrees to IIIdegrees in the progress,there are 12 genes(SNA I2,WISP2,M XRA 5,COMP,PRLHR,GA BRQ,SLC18A2,SIX2,IGKC,IGLC2,IGHG2,IGHG1)appear at the same time.(4)The late stage of POP happen,that there are 4 genes(WNT16,KCNG 1,ZCCHC12,IGHM)from II degrees to III degrees,III degrees IV degrees to progress could play an important role.Conclusion: The pathophysiology of POP is complex,including neurotrans mitter signaling pathway and Wnt signal transduction in POP had close contact.t.Differences between closely related and pelvic organ prolapse expression of candidate genes 10 genes may play a key role in the process of POP,and can beconsidered as a diagnosis of POP marker.
