Objective:To investigate the expression of PCDH10 in bladder urothelial carcinoma tissues and its relationship with clinicalpathology features as well as patients' prognosis,so as to provide theoretical basis and research base for the diagnosis,assessment of malignant behavior of bladder cancer and patients' prognosis.To investigate the promoter methylation status of PCDH10 in bladder urothelial carcinoma tissues,and correlated with its expression status in the first part of the study,so as to explore the correlation between PCDH10 methylation and its expression;then study the relationship between PCDH10 methylation and the development of bladder urothelial carcinoma as well as patients' prognosis.To investigate the promoter methylation status of PCDH10 in serum of patients with bladder urothelial carcinoma,and correlated with methylation status in the second part of the study,so as to explore the correlation between PCDH10 methylation in bladder urothelial carcinoma tissues and in serum of patients with bladder urothelial carcinoma;then study the relationship between PCDH10 methylation in serum of patients with bladder urothelial carcinoma and the development of bladder urothelial carcinoma as well as patients' prognosis.Methods:The expression of PCDH10 in 33 normal bladder mucosa and 105 bladder urothelial carcinoma tissues was detected using immunohistochemical study,and correlated with tumor number,size,shape,recurrence,grade,stage and patients' age,gender and prognosis.The methylation status of PCDH10 in 105 bladder urothelial carcinoma and 33 normal bladder mucosa tissues was detected by Methylation-specific PCR,and then associated with its expression status so as to illustrate the relationship between PCDH10 methylation and its expression,next correlated PCDH10 methylation with clinicalpathology features.The methylation status of PCDH10 in serum of 105 patients with bladder urothelial carcinoma and 33 normal control groups was detected by Methylation-specific PCR,and then associated with methylation status so as to illustrate the relationship between PCDH10 methylation in tissues and PCDH10 methylation in serum,next correlated PCDH10 methylation in serum with clinicalpathology features.Results:(1)The expression of PCDH10 was reduced in bladder urothelial carcinoma tissues compared with normal bladder mucosa tissues and the difference was statistically significant.Decreased expression of PCDH10 was correlated with tumor growth,recurrence,poorly differentiated and tumor invasion closely,but not associated with tumor number,shape or age and gender of the patients.In addition,the 5-year overall survival rate of 38 patients with PCDH10 expression normally was89.5%(34/38),but the 5-year overall survival rate of 67 patients with receded PCDH10 expression was 64.2%(43/67),the difference was statistically significant.(2)The methylation rate PCDH10 in bladder urothelial carcinoma tissues was 61.9%,however no methylation was found in normal bladder mucosa tissues,and the difference was statistically significant.PCDH10 methylation was negatively correlated with its expression in bladder urothelial carcinoma tissues.In addition,PCDH10 methylation was correlated with clinicalpathology features.(3)The methylation rate PCDH10 in serum of patients with bladder urothelial carcinoma was 55.2%,however no methylation was found in normal control group,and the difference was statistically significant.PCDH10 methylation in serum was positively correlated with its level in bladder urothelial carcinoma tissues.In addition,PCDH10 methylation in serum was correlated with clinicalpathology features.Conlusion: Gene PCDH10 promoter methylation is associated with the low expression of PCDH10,which reflected from that patient with gene PCDH10 promoter methylation in bladder carcinoma tissue accur the phenomenon of lower expression;The low expression of PCDH10 and gene PCDH10 promoter methylation in bladder tissues and in the serum were related to the occurrence of bladder urothelial carcinoma;PCDH10 low expression and gene PCDH10 promoter methylation in bladder tissue and the serum are associated with malignant behavior of bladder carcinoma,including the morphology,the size of the tumor recurrence and extent of differentiation and invasion depth;and low expression of PCDH10 and gene PCDH10 promoter methylation in bladder tissue and in serum can be used as independent judgment factors for the prognosis of bladder urothelial carcinoma.
