Cardiovascular disease,cerebrovascular disease,and diabetes mellitus have become the major threat to human public health.They are complications.Shu-nao-xin dropping pill is derived from a traditional recipe,which is composed of 2 herbs,Chuanxiong(Ligusticum chuanxiong Hort,Umbelliferae)and Danggui(Angelica sinensis radix,Umbelliferae).The main clinical indication of Shu-nao-xin dropping pill is cerebrovascular diseases like as migraine.At present,there is still no research on the modern pharmacology and substance bases of Shu-nao-xin dropping pill.Therefore,this thesis aims to study the cardiovascular protective effect and type 2 diabetes intervention and the substance bases of Shu-nao-xin dropping pill systematically.Antihypertensive,neuroprotective,and antidiabetic effects of Shu-nao-xin dropping pill were studied.The present reslults displayed that(1)Shu-nao-xin dropping pill could induce endothelium-dependent and endothelium-independent vascular relaxation in vitro and improve myocardial and aortic morphology abnormalities caused by hypertension in vivo.(2)Shu-nao-xin dropping pill could improve the learning and memory of D-galactose-induced aging rat model,which was associated with antioxidant stress,inhibition of neuron apoptosis and improvement of synaptic connections.(3)Shu-nao-xin dropping pill could reduce the blood glucose of type 2 diabetic rats.The hypoglycemic effect of Shu-nao-xin dropping pill might be related to anti-oxidative stress,regulate lipid metabolism and inhibit inflammatory response.The chemical constituents of Shu-nao-xin dropping pill were analyzed on the basis of clarifying the pharmacological effect of Shu-nao-xin dropping pill.The present results indicated that there were 15 constituents identified in Shu-nao-xin dropping pill.Four parent compounds including ferulic acid,senkyunolide I/H,senkyunolide A,and ligustilide were identifided in rat plasma.Two parent compounds including senkyunolide I/H and ligustilide were identifided in rat brain tissue.The pharamcological activities of ferulic acid and total lactone derived from Shu-nao-xin dropping pill were further evaluated based on the chemical composition and serum pharmacochemistry analysis.Ferulic acid and total lactone showed acute antihypertensive and neuroprotective effects.In addition,total lactone could reduce blood glucose in type 2 diabetic mice.The hypoglycemic effect of total lactone might be related to anti-oxidative stress,regulate lipid metabolism,inhibit inflammatory response and anti-islet cell apoptosis.These results suggested that ferulic acid and total lactone might be the main bioactive components in Shu-nao-xin dropping pill.In conclusion,all these research provide an experimental and theoretical basis of the therapy of cardiovascular disease and diabetes of Shu-nao-xin dropping pills.In addition,these researches relatively understand the substance bases and supply the basis for the further development of Shu-nao-xin dropping pill.
