【Objective】1.To investigate 5-ASA in prevention mechanism of caneration in ulcerative colitis2.To analyze the efficay of single vs multiple doses of 5-ASA in the treatment of mild-moderate ulcerative colitis3.To investigate the mechanism of targeting apoptosis-related proteins in inhibiting colon cancer cell proliferation and promotes apoptosis4.To investigate the incidence of psychological anxiety and depression in mild-moderate ulcerative colitis【Methods】I.To analyze the mechanism of 5-ASA in preventing caneration in ulcerative colitis1.To discuss the epidemiology of ulcerative colitis related colorectal cancer2.To discuss the chemoprophylaxis of mesalazine in preventing ulcerative colitis related colorectal cancerII.To analyze the efficay of single vs multiple doses of 5-ASA in the treatment of mild-moderate ulcerative colitis1.To collect 210 patients of mild-moderate ulcerative colitis from Oct.2013 to Aug.2015 from Shanghai Tenth’s Hospital and The First Affiliated Hospital of Kunming Medical University2.Visits were scheduled at week 0,2,4,and 6（final visit）,and vital signs,laboratory tests（haematology,biochemistry,urinalysis）and clinical signs were recorded at each visit.3.Endoscopy and histology were compared between baseline and the final visits（wk 6）.Colonic biopsies were obtained at baseline and final visits for evaluation of inflammatory cytokines by qRT-PCR.III.To investigate the mechanism of inhibiting colon cancer cell proliferation and promotes apoptosis by down-regulation of lncRNA CCAT11.Colon cancer tissue from ulcerative colitis associated colorectal cancer（UC-CRC）and matched adjacent non-tumor tissues（>2 cm from the tumor）were obtained from 7colon cancer patients who underwent surgery in Shanghai Tenth’s People’s Hospital,Tongji University,Shanghai,China from January,2010 to June,2015.The samples were snap-frozen in liquid nitrogen immediately.2.The colon cancer cell lines（SW620,HCT116 and HT29）and colon cell line（293-T）were cultured in DMEM.3.Deproteinizing RNA,detection of qRT-PCR,lncRNA PCR sequencing,Western blot analyzing,cell counting kit-8（CCK-8）and flow cytometry4.To analyze by SPSS 18.0IV.The Incidence of psychological anxiety and depression is highly increased in Ulcerative Colitis patients1.To collect the ulcerative colitis patients who visited the department of Gastroenterology,the 10^th Hospital of Shanghai,Tongji University,Shanghai,China between July 2015 to September 2015 were consecutively enrolled.In thisobservational study,patients with UC（n=65;observation group）and healthy volunteers（n=88;control group）were enrolled2.To collect social information and clinical data3 All participants completed the questionnaires of Self-Rating Anxiety Scale（SAS）and a Self-Rating Depression Scale（SDS）4.To analyze social-demographic characteristics and clinical data of those patients by SPSS 14.0【Results】1.Mesalazine is helpful to prevent caneration of ulcerative colitis.The mechanism are removing nitrogen metabolism,regulating the cell cycle and effecting the cellular pathways2.5-ASA 3g OD is efficacious to induce the clinical and endoscopical remission ofmild-moderate ulcerative colitis.Proinflammatory cytokines,C reactive protein（CRP）and erythrocyte sedimentation rate（ESR）were significantly decreased after treatment and histological grading was markedly improved after 3 g OD 5-ASA treatment3.lncRNA CCAT1 is partly related with the generation and progressing of tumorcell.It is highly expressed in colon cancer.Low lncRNA CCAT1 could inhibit the proliferation and promote the apotosis of colon caner cell.4.The incidence of psychological of anxiety and depression is highly increased in ulcerative colitis.It is related with personal characteristics and clinical types.Socio-demographic variables had no statistical difference between the ulcerative colitis group and control group.【Conclusions】1.5-aminosalicylic acid has partly effective to prevent the canceration of ulcerative colitis.2.3g OD of mesalazine is effective in inducing clinical and endoscopic remission of mild-moderate ulcerative colitis.Its adverse reaction is lower than 1g TID while the patients have better compliance.3.Targeting apoptosis-related proteins is effective in down-regulating lncRNA CCAT1 which could inhibits colon cancer cell proliferation and promotes Apoptosis4.The incidence of psychological of anxiety and depression is highly increased inulcerative colitis.It is related with personal characteristics and clinical types.Besides the regular treatment,psychological screening and counseling are essential to those patients.
