| Aim:To investigate the prognostic value of E-cadherin(ECD)expression in laryngeal cancer.Methods:In a clinical study,a total of 79 laryngeal cancer patients who received surgery were included in this study.The expression of ECD in laryngeal cancer tissue was detected by immumohistochemical staining.In an in vitro study,ECD up-regulated cells(Ad-ECD)and ECD down-regulated cells(sh-ECD)were cultured.Cell apoptosis was detected by flow cytometry.In an in vivo study,nude mice were injected with Ad-ECD,sh-ECD,and Hep-2 cells.The ECD expression and tumor metastasis in the brain,liver,and lung tissues were detected,and tumor metastasis was evaluated.Results:Clinical data showed that expression of ECD was statistically correlated with survival rate(p<0.05),but not with other clinical characteristics,such as age,gender,lymphatic metastasis,and TNM staging.ECD was down-regulated in sh-ECD cells and the down-regulation significantly reduced apoptosis of Hep-2 cells.Tumor growth was significantly inhibited in the Ad-ECD group.Except the tumor tissue,no positive ECD expression or significant tumor cells were found in the brain,liver,and lung tissues.Conclusion:Down-regulation of ECD may associate with infiltrative tumor growth and result in poor prognosis.However,ECD expression is not significantly associated with tumor metastasis.Aim:Screening and analyzing the difference of the mRNA expression spectrum of laryngeal cancer and nearby normal laryngeal mucosa to further explore the association of the genes related epithelium-mesoderm transition(EMT)with the invasion,metastasis,and prognosis of the laryngeal cancer.Method:Six pairs of fresh frozen tissue were collected and tested by human mRNA microarray.We use Jinxin biologic array general labeling kit to amplifying and immuno-labling of total RNA,then we analyze the hybrid images and extracted data by Aglient feature extraction software(vl0.7).Finally we use Aglient genespring software to calculate the difference of the gene expression level and statistically significant P value.If the difference of the expression level exceeds 2 fold and p<=0.05,the genes were considered as differentially expressed.Results:We got 2253 differentially expressed mRNA by array screening.Among them,1014 mRNA expression level unregulated,while 1149 down regulated.We further analyzed the data by multiple informatical database including KEGG pathway,PID,Biocarta,Reactome,Biocyc,PANTHER,GO,etc.and found that the top relationship pointing to the signaling pathway of the extracellular matrix.We further analyzed the EMT-related genes such as LAMC2(laminin gamma 2),P-cadherin,ZEB2(zinc finger E-box binding homobox 2),E-cadherin,N-cadherin,beta-catenin,vimentin,fibronectin,and found that only LAMC2 and P-cadherin were differential expressed genes.Conclusion:1.The gene array method is feasible to screen typical expression profile of laryngeal cancer and EMT-related genes.2.LAMC2 and P-cadherin are the potential new specific markers of EMT-related gene of the laryngeal cancer.Aim:Epithelial to mesenchymal transition(EMT)allows neoplastic cells to gain the invasive phenotype and produces single migratory cells,which are required for cancer progression and metastasis.In the present study,we investigated the expression of EMT-related biomarkers and their relationship with clinicopathological parameters in pharyngeal and laryngeal squamous cell carcinoma(PLSCC).Methods:E-cadherin,N-cadherin,P-cadherin,β-catenin,LAMC2(laminin gamma 2)and ZEB2 protein expression was evaluated with immunohistochemistry in a cohort of 72 patients with operable PLSCC who were treated between February 2007 and November 2013 at the Ear,Nose,and Throat(ENT)Department of Drum Tower Hospital,affiliated with Nanjing University,Nanjing,China.We analyzed the relationship between these transition markers and clinicopathological parameters and evaluated their prognostic impact in PLSCC.Results:Immunohistochemical analysis indicated that E-cadherin,N-cadherin,P-cadherin,β-catenin,LAMC2 and ZEB2 expression were differentially expressed in PLSCC and non-neoplastic tissues(P<0.05).Positive LAMC2 expression was correlated with later T stage(P<0.05).P-cadherin was non-significantly correlated with all clinicopathological parameters.Positive ZEB2 expression was correlated with later pathologic stage,lymph node metastases and worse tumor differentiation(P<0.05).Negative E-cadherin expression were correlated with later pathologic stage and lymph node metastases(P<0.05).Positive N-cadherin was correlated with later T stage,later pathologic stage and worse tumor differentiation(P<0.05).Positiveβ-catenin expression was correlated with later pathologic stage,worse tumor differentiation,lymph node metastases(P<0.05).Univariate analysis showed that E-cadherin,N-cadherin,β-catenin and ZEB2 were significantly correlated with OS while LAMC2 and P-cadherin were non-significantly correlated with OS.However,the co-expression of them and the other four biomarkers were significantly correlated with OS(P<0.05).In the multivariate analysis,it revealed that pathologic stage and positive expression of N-cadherin,β-catenin and ZEB2 were independent risk factors for OS in PLSCC using the Cox proportional hazards model.Conclusions:EMT-related proteins are differentially expressed in PLSCC and adjacent non-neoplastic laryngeal tissue.EMT mediates tumor progression and shortens overall survival(OS)in PLSCC.The co-expression of LAMC2 and P-cadherin and the other four biomarkers were significantly correlated with OS.Pathologic stage and positive expression of N-cadherin,β-catenin and ZEB2 could be independent predictors of prognosis in PLSCC. |
