| ObjectiveThis study was to observe the clinical efficacy and safety of fire needle combined with acupuncture in the treatment of chronic fatigue syndrome(CFS),which provide new ideas and basis for the treatment of CFS with fire needles.The animal experiment evaluates the efficacy of fire needle,acupuncture,and moxibustion in the treatment of CFS rat model,discusses the mechanism of antioxidation and immune function.It provides new ideas and objective basis for the treatment of CFS by acupuncture and moxibustion.MethodSixty-four patients with chronic fatigue syndrome were randomly divided into fire needle combined with acupuncture group and simple acupuncture group.Fire needle and acupuncture treatment were performed four weeks.The clinical symptom scores,FS-14 scores,self-rating anxiety scale(SAS),effective were tested to observe the efficacy and safety of fire needle combined with acupuncture in treating chronic fatigue syndrome.A total of 50 male SD rats were randomly divided into control group,model group,fire needle group,acupuncture group and moxibustion group.CFS model was established by bounding and swimming in cold for 14 days.Rats of fire needle,acupuncture group and moxibustion group were treated of fire needle,acupuncture and moxibustion acupoint “Gaohuang”(BL43)for 7 days.Before and after treament,the behavioral open field test,exhausted swimming time,and tail suspension time of the five groups of rats were recorded.The activities of SOD,GSH-Px and the levels of MDA,IL-1?,IFN-?,TNF-? in serum were elevated.To evaluate the efficacy of acupuncture in the treatment of CFS rat models.Results(1)The clinical randomized controlled trials showed that: There was no statistical difference in the CFS clinical symptom quantification scores,fatigue scale-14 scores(FS-14),and anxiety self-rating scale(SAS)scores between the baseline comparison treatment group and the control group(P>0.05),indicating that there was no difference in baseline scores between the two groups.The treatment group and control group compared with baseline,there was statistically difference in CFS clinical symptoms quantitative score,fatigue scale-14 scores(FS-14),anxiety self-assessment(SAS scores)(P<0.05).Which indicating that the treatment groupand control groupan improve the clinical symptoms of fatigue and anxiety.There was no significant difference in the quantitative scores of CFS clinical symptoms,fatigue scale-14 score(FS-14)and anxiety self-rating scale(SAS)scores between the treatment group and the control group at 2 weeks(P>0.05),and which were statistical differences at 4 weeks(P<0.05).The efficacy of the treatment group was better than the control group after 4 weeks of treatment.Compared the clinical efficacy of the two groups,0 was cured in both groups.In the treatment group,6 cases markedly effective(20.0%),23 cases effective(76.7%),1 case invalid(3.3%),the total efficiency is 96.6%.In the control group,1 case markedly effective(3.3%),21 cases effective(70.0%),8 cases invalid(26.7 %),the total efficiency is 73.3%,the total effective rate of the two groups showed a statistical difference(P <0.05),indicating that the fire needle combined with acupuncture can improve the clinical efficacy of treatment of CFS.There were no adverse events in the two groups of patients during the entire treatment process,indicating that fire needle combined with acupuncture treatment of CFS is safe and reliable.(2)Effects of acupuncture and moxibustion on the body mass and behavior of CFS model rats: Before the model was established,the body weight,exhaustive swimming time,tail suspension time and the open field of the rats of the fire needle group,the acupuncture group,the moxibustion group,the model group and the normal group.There was no significant difference in test scores,(P>0.05),indicating that the basic situation of the five groups of rats were the same.Before treatment,Fire needle group,acupuncture group,moxibustion group,model group and normal group compared rat's body weight,open field test score,exhausted swimming time,tail suspension time,the difference was statistically significant(P< 0.05).There was no significant difference in the body mass and behavior of CFS model rats between the fire needle group,acupuncture group,moxibustion group,and model group(P>0.05).The success of modeling was achieved,and there was no difference in the body mass and behavioral indexes between the four groups of the fire-acupuncture group,the acupuncture group,the moxibustion group and the model group.After the treatment,the body mass and behavior of CFS model rats of the fire needle group,acupuncture group,and moxibustion group and the model group were statistically significant(P<0.05),indicating that the fire needle group,acupuncture group,and moxibustion group can effectively improve the body mass of CFS rats and relieve the CFS model.After treatment,the body mass and behavior of CFS model rats of fire needles,acupuncture,moxibustion and normal groups were statistically significant(P<0.05).There was no difference of the body mass and behavior of CFS model rats between the fire needle group,acupuncture group,and moxibustion group(P>0.05),indicates that the fire needle group,acupuncture group and moxibustion group have no difference in the effect on the body mass and behavior of CFS model rats.(3)The effect of acupuncture and moxibustion on serum anti-oxidation indexes in rats of CFS model: Compared with fire needle group,acupuncture group,moxibustion group and normal group,the activity of SOD and GSH-Px in serum of model group rats were decreased(P<0.05).MDA levels increased(P<0.05),indicating that the serum levels of SOD and GSH-PX in CFS rats decreased and the levels of MDA increased,suggesting that the antioxidant capacity of CFS rat models was lower than that of normal rats.And the fire needle,acupuncture,moxibustion cream acupoints can improve the antioxidant capacity of CFS rats.There was no significant difference in serum SOD activity,GSH-Px activity and MDA level between the three groups of fire needle group,acupuncture group and moxibustion group(P>0.05),indicating fire needle group,acupuncture group,moxibustion Effect of three groups of treatment on serum SOD activity,GSH-Px activity and MDA level in rats No difference in loudness.(4)The effect of acupuncture and moxibustion at acupoints on serum cytokines in CFS model rats:Compared with fire needle group,acupuncture group,moxibustion group and normal group,serum IFN-? and TNF-? in model group were all decreased(P< 0.05),IL-1? levels were elevated(P<0.05),indicating that the CFS model group rats compared with the normal group,the body's inflammatory response increased,immune function decreased,and fire needles,acupuncture,moxibustion cream points can enhance CFS The immune function of the rats was inhibited and their inflammatory responses were inhibited.Fire needle group,acupuncture group,moxibustion There was no significant difference in the serum levels of IL-1?,IFN-? and TNF-? between the three groups(P>0.05),indicating that the three groups of fire needle group,acupuncture group and moxibustion group were There was no difference in the effects of serum levels of cytokines IL-1?,IFN-?,and TNF-? in CFS rats.ConclusionThe results of clinical trials in this study suggest that the combination of fire needles and acupuncture in the treatment of CFS can reduce fatigue,anxiety scores,and improve clinical symptoms.The clinical efficacy of fire needle combined with acupuncture in the treatment of CFS is better than simple acupuncture.This shows that fire needle combined with acupuncture treatment of CFS is effective,safe and reliable.The results of animal experiments preliminary showed that fire needle,acupuncture,moxibustion can effectively improve the body mass,physical and psychological fatigue of CFS rats,and can increase the body's antioxidant capacity,inhibit inflammation and enhance immune function in CFS rats.However,there are no significant differences in the efficacy,behavioral indicators,serum anti-oxidation parameters and cytokines of different acupuncture methods: fire needle,acupuncture,moxibustion.Therefore,it is indicated that fire needle,acupuncture,and moxibustion are effective methods for treating CFS model rats.Antioxidation and regulation of inflammatory reaction and immune function may be one of the mechanisms of treatment of CFS model rats.Different acupuncture methods treat CFS model rats,which show no significant difference in the antioxidation and immune function. |
