| Objective:To investigate the expression of Mel-18,Bmi-1,CD34 and D2-40 are in esophageal squamous cell carcinoma tissue and their clinical significance.Methods:Clinical data and the surgical specimens of 76 patients with esophageal squamous cell carcinoma were collected.The expression of Mel-18 and Bmi-1 in esophageal squamous cell carcinoma tissues and pericarcinous tissues were measured by real-time PCR and Western blotting.Expressions of Mel-18,Bmi-1,D2-40 and CD34 were detected in 76 cases of esophageal squamous cell carcinoma and 76 cases of adjacent nomal tissues using immunohistochemistry and tissue microarray.The MVD marked by positive CD34 and LVD marked by positive D2-40 were calculated.The Mel-18,Bmi-1,CD34 and D2-40 expression status and their correlationship in esophageal squamous cell carcinoma tissue,and the relations of their expressions with the clinicopathologic factors were analyzed.Results:The real-time PCR revealed that the negative expression of Mel-18 mRNA was 53.94%(41/76)in esophageal squamous’ cell carcinoma tissue.the positive expression of Bmi-1 mRNA was 63.16%(48/76)in ESCC.Western blotting revealed that the levels of Mel-18 was significantly lower expression(0.683±0.125)in ESCC tissue.The significantly higher expression of Bmi-1(0.985±0.104)in ESCC in comparison with nomal tissue.The immunohistochemical results revealed that the positive expression rate of Mel-18 was 30.3%(23/76)in esophageal’squamous cell carcinoma tissues,significantly lower than the rate 67.1%(51/76)in comparison with the normal esophageal tissue.The positive expression rate of Bmi-1 was 77.6%(59/76)in esophageal squamous cell carcinoma tissues,significantly higher than the rate 27.6%(21/76)in the normal esophageal tissue.The positive expression rate of CD34 was 88.2%(67/76)in esophageal squamous cell carcinoma tissues,significantly higher than the expression rate 31.6%(24/76)in the normal esophageal tissue.The D2-40 positive expression rate was 80.3%(61/76)in esophageal squamous cell carcinoma tissues,significantly higher than the expression rate 23.7%(16/76)in the normal esophageal tissue.The expression of Mel-18 has a correlation with lymph node metastasis,clinical stage,and depth of invasion,and not correlated with the patients’ tumor differentiation,age,sex.The expression of Bmi-1 was significantly correlated with clinical stage,lymph node metastasis and differentiation degree,and not correlated with the patients’ depth of invasion,age,sex.The MVD marked by positive CD34 in esophageal squamous cell carcinoma has a correlation with the differentiation degree,lymph node metastasis,clinical stage and depth of invasion,and not correlated with the patients’ sex and age.The LVD marked by positive D2-40 in esophageal squamous cell carcinoma has a correlation with depth of invasion,the differentiation degree,clinical stage and lymph node metastasis and not corr,elated with the patients’ age and sex.There has a negative correlation between the Mel-18 and Bmi-1 expression in esophageal squamous cell carcinoma by immunohistochemistry(r=-0.261,p=0.007).There has a positive correlation between The MVD marked by positive CD34 and LVD marked by positive D2-40 in esophageal squamous cell carcinoma by immunohistochemistry(r= 0.426,p=0.002).Conclusion:The Mel-18、Bmi-1、CD34 and D2-40 are correlated to the clinicopathological factors of the malignancy,and may play an predictive role in the development and progression of ESCC. |
