Ferns are widely used in the field of Chinese herbal medicine.Over a period of time,more and more attention has been paid to the development of new drugs around the world,especially to medicinal ferns.Among many ferns,Athyrium monkeyi,as a Chinese herbal medicine growing in Changbai Mountains,is an important fern that can meet both medical and edible needs.Relevant studies have shown that this medicinal fern can play an antioxidant role and inhibit the growth of bacteria.However,there is no systematic study on the anti-inflammatory effect of pteridophyte,Athyrium monkey-leg,at home and abroad.In order to investigate the anti-inflammatory effect of ethanol extract of Athyrium erinaceus on LPS-induced lung injury deficiency in mice and the effect of ethanol extract on LPS-stimulated peritoneal macrophages in mice,the effects of ethanol extract of Athyrium erinaceus on the expression of key proteins and inflammatory factors were investigated by means of signal transduction pathway platform.The anti-inflammatory and immune effects of pteridophytes are explained scientifically and reasonably.which can provide a reliable theoretical reference for the development and utilization of Pteridophyta monkeys.The toxic effects of ethanol extract from Athyrium hypoglaucum on peritoneal macrophages in mice were analyzed by MTT method;the effects of Athyrium hypoglaucum on the secretion of TNF-alpha and IL-6 induced by LPS were investigated by ELISA method;and the effects of key protein molecules related to inflammation signaling pathway were studied by Western blot method.Through careful study,it was found that when the concentration of ethanol extract from Athyrium monkeys was at 0-200.0 ug/mL,it did not poison the macrophages in the abdominal cavity of mice;at the same time,it significantly hindered the secretion of TNF-alpha and IL-6;in addition,ethanol extract from Athyrium monkeys could effectively regulate the inflammatory signaling pathway and play an anti-inflammatory role.In addition,the effects of ethanol extract from Athyrium hypoglossus on LPS-induced inflammation pathway and the protective function of ethanol extract on acute lung injury in mice were also discussed.In the course of the experiment,the author set up blank control,LPS induction,Pteris hypogaea(5.0 mg/kg body weight,10.0 mg/kg body weight)+LPS and dexamethasone(7.8 mg/kg body weight)+LPS and other groups of experiments.Ethanol extract of Pteris hypogaea was injected into the stomach of mice for 8 days,and dexamethasone was injected into the abdominal cavity of mice two hours before LPS induction began.In mice.After that,the lungs of mice were taken out to form alveolar lavage fluid,and the specific number of inflammatory cells in the lavage fluid was monitored.The levels of inflammatory mediator NO were detected by Griess method.Combined with ELISA method,the levels of TNF-alpha,IL-lbeta and IL-6 were obtained.Pathological sections were made to study the histopathological changes.The results showed that the ethanol extract of Athyrium mongolicum could significantly reduce the content of inflammatory cells in alveolar lavage fluid of mice,and inhibit the production of NO,TNF-a,IL-lbeta and IL-6.The conclusion showed that the ethanol extract of Athyrium mongolicum had significant anti-inflammatory effect on LPS-induced acute lung injury model in mice.In vivo antioxidant experiment,monkey legs athyriaceae fern extract,petroleum ether and dichloromethane layers to remove DPPH and ABTS effect is best,petroleum ether layer in 0.2 mg/mL clear effect DPPH reached 87.33%,at 0.2 mg/mL,the clearance of ABTS reached 98.73%,acetic ether layer of iron ion chelating ability best,ethyl acetate in 10 mg/mL,chelating ability by 75%.The following conclusions were drawn from the study:1.Extracts of brachiopteris simian might resist the inflammatory effect of LPS on mice by inhibiting the production of inflammatory factors through the dual pathways of MyD88 and TRIF;2.The extract layer of ethyl acetate of brassica monksholica had the best anti-nitric oxide ability,the extract layer of petroleum ether and dichloromethane had the best effect of removing DPPH and ABTS,and the extraction layer of ethyl acetate had the best ability of iron ion chelating.
