Intra-cerebroventricular Injection Of Anti-somatostatin-serum Involved In The Inflammatory Pain And Electro-Acupuncture Analgesia In Rats

Objective:To investigate the influence of endogenous somatostatin(SS) on the FAC-indced inflammatory pain and acupuncture analgesia,to investigate their possible central mechanism.Methods:AA rats were used as the subjects of the study,by using methods of behavioral pain threshold test,radioimmunity, ABC-ELISA, HPLC-ECD, immunohisto- chemistry,and Western blot,we observed AA rats’pain threshold ,the level of IL-6 and IL-10 in blood serum,the express of SS,the content of 5-HT andβ-EP in hypothalamus and spinal cord,and the express of Gi in hypothalamus after icro-injection of ASSS into lateral cerebral ventricles. Observe the changes of pain threshold after injection anti serum of 5-HT andβ-endorphin on anti serum of SS or electro-acupuncture.Results:Using icro-injection of ASSS into lateral cerebral ventricles ,rats’pain threshold can be reduced,the level of IL-10,the level of 5-HT andβ-EP in hypothalamus and spinal cord ,the express of SS in hypothalamus also can be reduced,while the level of IL-6 and the express of Gi-protein in hypothalamus can be increased. If adding injection the anti-serum of 5-HT and the anti-serum ofβ-EP ,the pain threshold mostly decreased than before, and the expression of SS in hypothalamus decreased than before.Conclusion:The SS may be an important regulative point on inflammatory pain and acupuncture analgesia. The SS may be related to the 5-HT orβ-EP and the IL-6 or IL-10 during the process of electro-acupuncture analgesia.Endogenous SS may be an important regulative point for acupuncture analgesia and regulation of NEI. The central mechanism may be related to the Gi-protein mediated signal transduction pathway.
