ObjectiveSeveral recent genome-wide association studies(GWAS)have linked SCN10A and SCN5A to PR interval,QRS duration and atrial fibrillation(AF).This study aims to investigate the relationship between SCN10A and SCN5A polymorphisms and AF recurrence after catheter ablation in a Chinese Han Population.MethodsA total of 502 AF patients undergoing catheter ablation were enrolled in the present study.AF recurrence was defined as a documented episode of any atrial arrhythmias lasting≥30s after a blanking period of 3 months.We collected peripheral blood from patients and extracted genomic DNA.A total of 15 tagging-single nucleotide polymorphism(t SNP)in SCN10A and 6 t SNP in SCN5A were genotyped.A multivariable logistic regression analysis was performed to evaluate the association between genetic polymorphisms and AF recurrence within 12 months after ablation.Kaplan-Meier survival curves and the log-rank test were used to compare AF recurrence differences among different groups in the the long-term analysis.In addition,multivariate Cox regression analysis was performed to evaluate the relationship between t SNP and AF recurrence in the long-term analysis.Results1.There was a significant difference between the AF recurrence group and no recurrence group in the allele distribution(P=7.86×10-5)and genotype distribution(P=1.42×10-5)of SCN10A rs6795970.The AF recurrence rate of rs6795970 GG genotype was significantly higher than GA+AA genotypes(30.5%vs.12.6%,OR3.05,95%CI 1.84~5.05,P=8.4×10-6).In a multivariable logistic regression analysis,rs6795970 genotypes were significantly associated with AF recurrence in both the additive model(OR 0.38,95%CI 0.24~0.62,P=1.09×10-4)and the dominant model(OR 0.31,95%CI 0.18~0.53,P=1.66×10-5).AF recurrence was observed in 28.1%in the long-term follow-up.Kaplan-Meier survival curves showed that the recurrence rate of rs6795970 GG genotype was significantly higher than GA+AA genotypes(33.8%vs.16.6%,log rank P=0.0005).In the multivariate Cox regression analysis,rs6795970 genotypes were significantly associated with AF recurrence in both the additive model(HR 0.48,95%CI 0.33~0.70,P=1.6×10-4)and the dominant model(HR 0.42,95%CI 0.28~0.64,P=0.001).Cox regression analysis further showed that rs6795970 GG genotype(HR 2.36,95%CI 1.56~3.58,P=0.001)was an independent risk factor of AF recurrence in the long-term follow-up.In addition,there was no significant association between other 14 t SNP and AF recurrence.2.In the analysis of the relationship between 6 SNPs of SCN5A and AF recurrence within 12 months after ablation,there was no significant difference in the allele distribution and genotype distribution between the AF recurrence group and no recurrence group.Moreover,there was no significant association between 6 t SNP of SCN5A and AF recurrence within 12 months.In the the long-term analysis,there was no significant association between these t SNP and AF recurrence.ConclusionsThese data suggest that SCN10A rs6795970 polymorphisms are associated with AF recurrence after catheter ablation in the Chinese Han Population.The rs6795970GG genotype is a risk factor of AF recurrence.The rs67950970 polymorphisms may be a potential biomarker for risk asscessment of AF recurrence after catheter ablation.
