Objective Observe the effects of the rehmannia dride rhizome(sovereign drug of JIEDUQUYUZIYIN methods)in treating and preventing glucocorticosteroid-induced osteoporosis(GIO)in MRL/lpr mouse,and approach the mechanism at the view of that Bone Morphogenetic protein-7(BMP-7)is the interaction protein of the glucocorticoid receptor(GR),on the base of the conclusion that JIEDU QUYU ZIYIN methods can prevent and treat GIO in SLE.Methods ①Look for the protein which is not only the interaction protein of glucocorticosteroid receptor,the key of the glucocorticosteroid effect mechanism,but also has the effect in bone metabolism,by using two dimensional electrophoresis and mass spectrum scanning.②Once the interaction protein of GR is founded,the experiment of MRL/Ipr mice will carry on.27 MRL/Ipr mice were divided into 3 groups randomly,and 9 mice were in each group.Blank group(group I):physiological saline,0.2ml/10g·d,intragastric administration;model group(group II):fed with the suspension of prednisone,0.1 ml/10g·d(1mg/ml)+physiological saline,0.2ml/10g·d;rehmannia dride rhizome group(groupⅢ):fed with the suspension of prednisone,0.1 ml/10g·d(1mg/ml)+the water solution of rehmannia dride rhizome,0.1 ml/10g·d(0.25g/ml);radix paeoniae rubra group(groupⅣ):fed with the suspension of prednisone,0.1 ml/10g·d(1mg/ml)+the water solution of radix paeoniae rubra,0.1 ml/10g·d(0.25g/ml);spreading hedyotis herb group(group V):fed with the suspension of prednisone,0.1 ml/10g·d(1mg/ml)+the water solution of spreading hedyotis herb,0.1 ml/10g·d(0.25g/ml).After 8 weeks,take the left upper extremity,using the X-ray function of the little animal image formation system to detect the bone mineral density(BMD).Take the right upper extremity,then made it into the pathological section,and observe the pathogen’s image after HE coloratura.Take the bone of both lower extremities,and douche the bone marrow by using 1ml syringe.Extract the total protein,then detect the content of protein using the method of the WesternBlot.Results ①The result of mass spectrum scanning:There are more than 30 points in the gelatin of two dimensional electrophoresis,and point 14 was proved BMP-7,which is the protein we looked for.②Pathology of the bone trabeculae:the bone trabeculae of group Ⅰis thick,and aligned tidily;group Ⅱ is thin,disrupted and untidiness;group Ⅲ is thick,and aligned tidily;group Ⅳ is thin,and untidiness;group Ⅴ is thin,disrupted and untidiness.③Bone density:group I:4.10±0.48;group Ⅱ:2.99±0.32;groupⅢ:3.72±0.88;groupⅣ:3.43±0.83;group Ⅴ:3.20±1.27.Statistics analysis by using SPSS 16.0 softwear:group I compared to group Ⅱ,P=0.000,has the statistical significance;group Ⅱ compared to groupⅢ,P=0.033,has the statistical significance;group Ⅱ compared to groupⅣ,P=0.151,has no statistical significance;group Ⅱ compared to group V,P=0.64,has no statistical significance.④The result of interest protein in bone marrow detected by western blot:the gray scale of GR:gruop Ⅰ 24.90,gruopⅡ:22.13,gruopⅢ:28.60;the gray scale of BMP-7:gruop 10.60,gruopⅡ:1.44,gruopⅢ:2.00.Conclusion BMP-7 maybe one of the interaction proteins of GR;Lots of GR were activated when ponderosus glucocorticoid were administrated,then lots of BMP-7 binded to the GR resulting down regulating the activity of BMP-7.So the body secreted more BMP-7,but still can not redeem the content of BMP-7 losting activity,which induced osteoporosis;The rehmannia dride rhizome can prevent and treat the GIO of MRL/lpr mice.It may insterfere in the interaction between the GR and BMP-7,and promote the content of BMP-7 in the bone marrow,which enhance the Myeloid Stem Cells differentiation to osteoblast,so allivate the GIO.
