Background Laryngeal squamous cell carcinoma refers to a malignant tumor that occurs in the larynx.According to statistics from the World Health Organization,laryngeal squamous cell carcinoma is more common in males than females,with a male to female incidence ratio of approximately 4:1.In addition to gender,age is also an important risk factor.Laryngeal squamous cell carcinoma usually has a higher incidence in the age group over 40 years old,especially between 60 and 70 years old.Smoking and alcohol consumption are one of the main risk factors for laryngeal cancer.In addition,HPV infection may also increase the incidence rate of laryngeal cancer.Although the incidence rate of laryngeal cancer has declined in some countries,it is still a serious health problem worldwide and needs more attention and research.Long chain non-coding RNA is a kind of RNA that has been widely studied in recent years,and has been proved to play a key role in a variety of cancers.Therefore,the role of HAGLROS is of great significance in the occurrence and development of cancer,as it can serve as a potential therapeutic target for tumors and as a potential biomarker to assist in the diagnosis and prognosis evaluation of cancer.This may provide theoretical basis and new research directions for targeted treatment of laryngeal squamous cell carcinoma.Objectives We aim to reveal the expression of HAGLROS in laryngeal squamous cell carcinoma(LSCC)and its impact on malignant behavior.We also investigate the potential mechanisms of HAGLROS in promoting the malignant behavior of LSCC,and provide a theoretical basis for HAGLROS as a potential biomarker for LSCC.Methods We used quantitative real-time PCR to detect the expression of HAGLROS in tissue,serum,and LSCC cell lines of patients with LSCC and analyzed its correlation with clinical pathological indicators.By transfecting HAGLROS overexpression plasmids and sh HAGLROS,we further explored the function of HAGLROS and its effect on LSCC cell proliferation,apoptosis,and angiogenesis.We used Western blot to detect the expression of vascular endothelial growth factor(VEGF),proliferating cell nuclear antigen(PCNA),P27,and cleaved caspase-3,as well as their effects on peripheral signaling pathways such as Erk1/2,JNK,and AKT.Results The results showed that HAGLROS was highly expressed in laryngeal squamous cell carcinoma tissues and cells,and was associated with lymph nodes,tumor depth,and clinical staging in patients with laryngeal squamous cell carcinoma.This indicates that HAGLROS may play an important role in the occurrence and development of laryngeal squamous cell carcinoma.In addition,our experimental results also indicate that the proliferation ability of laryngeal squamous cell carcinoma cells is higher than that of Hu La PC cells,which provides a basis for our subsequent experiments.Next,we investigated the effect of HAGLROS on laryngeal squamous cell carcinoma cells.We investigated the biological function of HAGLROS by transfecting the HAGLROS overexpression plasmid and sh HAGLROS.The experimental results indicate that overexpression of HAGLROS promotes the proliferation and angiogenesis ability of laryngeal squamous cell carcinoma cells,while inhibiting apoptosis.On the contrary,silencing HAGLROS has the opposite effect in laryngeal squamous cell carcinoma cell lines,inhibiting proliferation and angiogenesis,and promoting apoptosis.In order to further investigate the mechanism of the effect of HAGLROS on laryngeal squamous cell carcinoma cells,we further tested the expression of some related proteins.The experimental results of overexpression of HAGLROS showed that it increased the expression of VEGF and PCNA by activating the Erk1/2 and AKT or JNK signaling pathways in laryngeal squamous cell carcinoma cells,and decreased the expression of P27 and shear cysteine protease-3.This indicates that HAGLROS promotes the proliferation and angiogenesis of laryngeal squamous cell carcinoma cells and inhibits apoptosis by regulating the expression of these proteins.Conclusion This study found that HAGLROS is highly expressed in the tumor tissue and cell lines of patients with laryngeal squamous cell carcinoma.The high expression of HAGLROS is related to lymph node metastasis,tumor depth,and clinical staging of laryngeal squamous cell carcinoma patients.Overexpression of HAGLROS can promote the proliferation and angiogenesis ability of laryngeal squamous cells,and reduce cell apoptosis,while silencing HAGLROS has the opposite effect.Overexpression of HAGLROS can activate the Erk1/2 and AKT or JNK signaling pathways,upregulate the expression of VEGF and PCNA,but downregulate the expression of P27 and Cleaved Caspase-3.This study provides a basis for further research on the therapeutic effects of HAGLROS in laryngeal squamous cell carcinoma.
