Objective:1.To known the willingness of catgut implantation at acupoint treatment from the perspective of AR patients, that is,the awareness, recognition and selection of catgut implantation at acupoint treatment for AR;It also tries to explore the trigger point that may be widely accepted by patients, paving the way for future clinical research.2.The effect of catgut implantation at acupoint on the regulation of intestinal microbes in AR rats was analyzed by catgut embedding at Yingxiang acupoint and16 S r NDA high-throughput sequencing in AR rats.Methods:1.Based on the theory of KABP design a good reliability and validity of the questionnaire,AR patients scanning We Chat qr code network questionnaire survey method was applied to investigate that popularity, recognition, and selectivity of the therapy of catgut implantation at acupoint, analysis on it in patients more than 14 with AR,in the clinic of the university affiliated hospital otolaryngology.2.Twenty-four healthy rats were randomly divided into three groups: the first group:healthy blank group(group A),the second group: AR model group(group B),and the third group: catgut implantation at acupoint group(group C).Then the AR rat model was established(group 2 and group 3),and catgut implantation at acupoint intervention was performed at Yingxiang acupoint(group 3).Subsequently,fecal samples of rats in each group were collected and fecal DNA was extracted.16 S r NDA sequencing of DNA amplification region V3V4 was performed on sequencing platform Miseq to analyze the regulatory effect of catgut implantation at acupoint on intestinal microflora of AR rats.Results:1.A total of 347 valid questionnaires were collected in the formal investigation of this study.(1)Awareness: the number of people who knew catgut implantation at acupoint therapy was 189(54.47%),more than half of the total number of respondents; There were 165 respondents who knew that catgut implantation at acupoint could be used for the treatment of allergic rhinitis, accounting for 87% of the patients who knew catgut implantation at acupoint.Ninety-eight(28.24%)patients had been treated with catgut implantation at acupoint for allergic rhinitis, accounting for 60% of the known it.(2)Recognition: 70% of respondents were willing to recommend catgut implantation at acupoint for the treatment of allergic rhinitis.(3)Degree of choose: 261 people were willing to voluntarily propose this therapy for the treatment of allergic rhinitis, and 273 people were willing to accept the doctor’s recommendation to use it for their AR,accounting for about 2/3 of the total number respectively.(4)For the choice of the form of catgut implantation at acupoint therapy,"according to the condition of the doctor to decide" accounted for 1/3,"according to the economic situation" accounted for 25.65%,"do a few times to see the effect and then decide" close to 1/3 of the total number of people investigated.(5)Correlation: The degree of catgut implantation at acupoint knowledge, recognition and selection did not show significant correlation with the general information and disease information of the respondents.(6)Respondents’ choice of catgut implantation at acupoint therapy for AR plays a role through the mediating variable M "whether they are willing to recommend catgut implantation at acupoint therapy to their family and friends"."Known the curative effect of catgut implantation at acupoint therapy" is not a bridge between "whether you know catgut implantation at acupoint therapy" and "whether you choose catgut implantation at acupoint therapy".2.(1)Alpha diversity: Compared with healthy rats, the total flora of AR rats changed significantly;after catgut implantation at acupoint treatment, the total number of intestinal flora in AR rats did not change significantly compared with healthy rats, but significantly changed compared with AR rats.(2)Beta diversity(at Phylum level and Genus level):(1)Analysis of phylum level NMDS: model group(group B)and blank group(group A)were well distinguished;catgut implantation at acupoint group(group C)and model group(group B)can be distinguished well. There were obvious proximity and overlap between group C and A(blank group).(2)Dominant phylum horizontal flora: There are dominant bacteria such as Firmicutes,Bacteroidetes,Proteobacteria and Actinobacteria in AR rat phylum level regulated by acupoint catgut.(3)Changes of phylum level flora content: catgut implantation at acupoint intervention up-regulated Deferribacteres in the intestinal phylum of AR rats, and significantly up-regulated Candidatus_Saccharibacteria and Tenericutes. It can down-regulate Acidobacteria.(4)NMDS analysis of genus level flora: Group B and group A could be well distinguished; There was a certain degree of differentiation between the acupoint embedding group(group C)and the model group(group B).Group C overlaps with group A.(5)Dominant bacteria of horizontal flora: There are also some dominant bacteria at the genus level, including Lachnospiraceae_unclassified,Ruminococcaceae_unclassified,Porphyromonadaceae_unclassified and Prevotella is more abundant.(6)Content of genus level flora: catgut implantation at acupoint intervention ran to completion of Lachnoclostridium,Oscillospira,Bacteria-Unclassifed,and Bacteroidetes at the intestinal level of AR rats_Unclassifed had an up-regulation effect; It significantly down-regulated Murimonas,Paraprevotella and Acetatifactor.(7)There were some positive and negative correlations among species at phylum level and genus level. For example,at phylum level,Firmicutes were significantly negatively correlated with Bacteroidetes(P<0.001).Proteobacteria were significantly positively correlated(P<0.001).At the genus level,Ruminococcaceae_unclassified was significantly negatively correlated with Oscillospira(P<0.001);There was a significant positive correlation between Acetivibrio and Bilophila(P<0.05).(3)Lefse analysis: There were 6 species with significant differences at phylum level in AR rats after catgut implantation at acupoint intervention.P_Actinobacteria,P_Bacteroidetes,P_Elusimicrobia,P_Firmicutes,P_Spirochaetes.and P_Tenericutes.Conclusion:1.(1)Within the scope of investigation, the awareness, recognition and choice of catgut implantation at acupoint treatment willingness of AR patients are well, while the data of a larger range of willingness survey need further study.(2)Within the scope of investigation, catgut implantation at acupoint therapy for AR has the potential for further clinical research. The trigger point of extensive clinical research is whether patients are willing to recommend catgut implantation at acupoint therapy to their family and friends.2.(1)catgut implantation at acupoint has a two-way regulatory effect on the intestinal flora of AR rats.catgut implantation at acupoint can promote the intestinal flora number of rats to normal state.(2)Catgut implantation at acupoint can regulate the level of phylum and the intestinal flora of AR rats at genus level is similar to that of healthy rats. There were some dominant bacteria at phylum level and genus level respectively.Catgut implantation at acupoint could regulate the abundance of some bacteria at phylum level and genus level in AR rats. There were some positive and negative correlation between phylum level and genus level in AR rats regulated by catgut implantation at acupoint.(3)There were significant differences in phylum level of AR rats regulated by catgut implantation at acupoint, such as Actinobacteria and Bacteroidetes.(4)The research direction of intestinal flora regulation of AR by catgut implantation at acupoints has certain research potential and can be further studied. | | Keywords/Search Tags: | Catgut Implantation at Acupoints, Allergic rhinitis, Intend to, Questionnaire survey, 16s rDNA,
