Objective(1)To retrospectively analyze the serum omentin-1 levels in SONFH patients and healthy people,and to explore the clinical value of omentin-1 in the early diagnosis of SONFH.(2)To investigate the effect of omentin-1 on the proliferation and osteogenic differentiation of Mc3t3-el cells.(3)To explore the effect of gynostaponin on the proliferation and osteogenic differentiation of Mc3t3-e1 cells based on omentin 1 regulation,so as to provide a scientific basis for the early diagnosis,potential therapeutic targets and prevention and treatment of SONFH with traditional Chinese medicine.Methods(1)Serum samples of 64 patients with SONFH and 64 healthy controls were collected from the sample bank.Serum omentin-1 concentration was detected by ELISA.The correlation between serum omentin-1 and clinical indicators(ARCO stage,VAS score and Harris score)was analyzed in SONFH patients.ROC curve was drawn to evaluate the accuracy of serum omentin-1 in the diagnosis of SONFH.(2)CCK-8 assay was used to detect the effects of Dex and omentin-1 on the proliferation activity of Mc3t3-el cells,and ALP activity assay was used to detect the effects of omentin-1 on the proliferation activity of Mc3t3-e1 cells induced by Dex.Rt-qpcr and Western Blot were used to detect the effects of omentin-1 on the mRNA and protein levels of Runx2,Osx and OPN.(3)CCK-8 assay was used to detect the effect of gypenoside on the proliferation activity of MC3T3-E1 cells,and ALP activity assay was used to detect the effect of gypenoside on the proliferation activity of MC3T3-E1 cells induced by Dex.Rt-qpcr and Western Blot were used to detect the effects of gypenoside on the mRNA and protein levels of Runx2,Osx,OPN,and omentin-1.ResultsThe serum omentin-1 concentration level of SONFH patients is decreased,and serum omentin-1 can be used as a potential serum biochemical marker for the early diagnosis of SONFH.Omentin-1 can improve the proliferation and ALP activity of osteoblasts induced by Dex,and promote osteogenic differentiation.Chinese medicine monomer gynostemma pentaphyloside can improve cell proliferation and promote osteogenic differentiation,and its mechanism may be through omentin-1 based regulation.ConclusionThe serum omentin-1 concentration level of SONFH patients is decreased,and serum omentin-1 can be used as a potential serum biochemical marker for the early diagnosis of SONFH.Omentin-1 can improve the proliferation and ALP activity of osteoblasts induced by Dex,and promote osteogenic differentiation.Chinese medicine monomer gynostemma pentaphyloside can improve cell proliferation and promote osteogenic differentiation,and its mechanism may be through omentin-1 based regulation.
