In this paper, ribosome 28S,mitochondrion cytochrome b (Cytb),cytochrome oxidase I(COI) and sequence—related amplified polymorphism(SRAP) were used to analysis the phylogenetic relationships,gene sequence,and intraspecific distance among Ficus pumila & Ficus awkeotsang pollinating wasps(W. pumilae), Ficus virens var. sublanceolata (Miq.) Corner pollinating wasps(Platyscapa coronata),unpollinating wasps(Camarothorax bimasculinus),Ficus altissima pollinating wasps(Eupristina altissima).The results were as follows:1. Ribosome 28S analysis result:Ribosome 28S fragments were amplified via polymerase chain reaction,recycled and purificated, the sequences analysis as followed:①The average content of A, G,T, C was 22.2%,30.08%,22.51%,25.20%, the average content of GC was 55.28% higher than AT;②The Kimura2-paramter distance(K2P) calculated by MEGA3.1,the average distance between the F.awkeotsang pollinating wasps (W.pumilae) was 0.0161, the average distance between the F. pumilae pollinating wasps(W.pumilae) was 0.0103, the average distance between the two group was 0.0532, the genetic distance between two different morphology C.bimasculinus male wasps of was 0.0069;③the clustering analysis with the method of NJ and UPGMA. The results were as follow:the variation between the W.pumilae via F.pumila and W.pumilae via F.awkeotsang mainly came from the inter-group, the genetic relationship between the two different C.bimasculinus male wasps was close, belong to male polymorphism. The analysis of NJ and UPGMA proved the above conclusion.2. The analysis results of Mitochondrion cytochrome b (Cytb) and cytochrome oxidase I(COI):(1)Cytb sequences analysis as followed:①the average content of A, G, T, C was 32.9%,11.2%,41.9%,14.1%, the average content of GC was 25.3% lower than AT; G-base was lower than the others and generally showed a lack of G-base;②gene sequence analysis showed that conserved site was 88.67% and variable site was 13.09%;③the average K2P distance between the F.awkeotsang pollinating wasps was 0.0244, the average distance between the F.pumila pollinating wasps was 0.0097,and the average distance between them was 0.1305;④analysis with the method of NJ and UPGMA.(2)COI sequences analysis as followed:①the average content of A, G, T, C was 31.5%,16.9%,40.6%,11.0%, the average content of GC was 27.9%,C-base was lower than the other especially on the third point;②gene sequence analysis showed that conserved site was 89.08% and variable site was 10.92%;③the average K2P distance between the F.awkeotsang pollinating wasps was 0.0068, the distance between the F.pumila pollinating wasps was 0, the average distance between them was 0.0956,was as 10 times as the two group separatly;④using NJ and UPGMA method to analyze.The figures showed that the relationship between them was stability and credible, pollinating wasps(Wiebesia pumilae) which lived in Ficus pumila and Ficus awkeotsang separately were cryptic species.3. SRAP were used to analysis genetic distance and genetic similarity among the experimental samples,17 pairs primers selected in 64 pairs primers could get 491 polymorphic bands. The bands amplified were between 100bp and 2000bp, the average genetic distance between the Ficus pumila and Ficus awkeotsang pollinating wasps(W.pumilae) was 0.210908; the genetic distance between the different morphology C.bimasculinus male wasps was 0.150713. The results showed that the genetic relationship between the W. pumilae via F.pumila and W.pumilae via F. awkeotsang was close,but the date showed that the intraspecific distance was no more than 0.2,and the species distance was between 0.2 and 0.8,so the pollinating wasps(W.pumilae) of the Ficus pumila and Ficus awkeotsang were already reach the degree of the species diversity; similarly, the genetic relationship between the different C. bimasculinus male wasps was close. The clustering result was consistent with the above conclusion.The results of the 28S,Cytb,COI,SRAP showed that the pollinating wasps(Wiebesia pumilae) which lived in Ficus pumila and Ficus awkeotsang separately were already reach the degree of species diversity, they were cryptic species;the different morphology C.bimasculinus male wasps were male polymorphism.
