Ciliate cortex are rich in cytoskeletal elements, microtubules are the main component of the cytoskeleton, cytoskeleton microtubules include cortical ciliature and cortical non ciliature microtubules. Cortical ciliatures microtubules mainly by the microtubules of cilia, the matrix, the matrix skeleton and small fibers; non ciliature microtubule reticular structure, form of the microtubule network, microtubule layer and microtubules, is structure to maintain cell shape and essential to ensure normal cell movement. Hypotrichia ciliate has a variety of ciliatures in the ventral cortex or dorsal cortex, cilia rod and the matrix is the structural basis for the arrangement according to the different composition and position, also this has a significant difference in the feature.Microtubule protein containing the basic components of the complex microtubule unit in such cell cortex,making the ciliate as good material in the research of distribution and function of associated proteins of the substrate and the microtubules. In recent years, the use of tubulin fluorescence labeling methods to explore the micro-organelles in ciliates and protein components of microtubules. micro-organelles function and mechanism of microtubule assembly, which has become a research hotspot in the field. Studies on the protozoan cytoskeleton, cell structure for understanding the diversity of the evolution of cells, the regulation of cellular activities, etc.,is of great scientific significance and potential applications. This technique of scanning electron microscopy, laser scanning confocal microscopy and transmission electron microscopy technique to target hypotrichia ciliate Oxytricha platystoma as materials, observed the structure of cell in ciliates cortex cytoskeleton, microtubules and cytoplasmic organelles at microscopic level, the results are as follows:1 Observation of ciliate cortex surface structure by SEMScanning electron microscopy technique showed that in Oxytricha platystoma ventral cortex, adoral zone of membranelles contain 39-47 pieces small membranes, most of the small membrane matrix with 4 columns. Features include 2 undulating membrane matrix, the matrix composed of cilia protruding upward long and wide diaphragm; the right of the membrane lining of AZM at the edge of port side AZM lining in the inner lining of the adoral zone; on the back of the ciliate, a total of six dorsal cirrus; The frontal cirri, ventral cirri and transverse cirri are distributed by 8-5-5 mode, left and right edge of cirri each one. Compared O. platystoma of Oxytricha and Urostyla grandis of Urostyla, hypotrichida Oxytricha ciliates cirri structure in adoral cortex and AZM have its specificity of Oxytricha. Compared with the same family of the genus Stylonychia and Tachysoma. The base-associated microtubules of the FVTC, L-and RMC contain anterior longitudinal microtubules, posterior longitudinal microtubules and transverse microtubules or radiate microtubules. The results showed that cortical ciliature base microtubules of O. platystoma have their species specificity, such as, UM of the O. platystoma form well-developed microtubule net at the base of the special long cilia series. The structure of such large AZM. UM and the base-associated microtubule skeleton have not been reported in ciliates. In addition, the oral-end bracket and postoral microtubules may be prevalent in hyportrichia ciliate base of adoral ciliature microtubules. Explore the structure of ciliate cell organelles and organelle positioning in the terms of features, for further understanding of cell organelles ciliates diversity and genetic structure, the function of organelles and cellular activities aspects of the relationship is meaningful.3 Observation of cortical microtubular organelles and cytoplasmic organelles by TEM By transmission electron microscopy technique observed Oxytricha platystoma structure of cell cortical cilia microtubules and cytoplasm organelles. The cell cortex underlying ciliary microtubular organelles in the device by the adoral cilia organelle microtubules, ciliary body and ciliary base-associated microtubules. These structures as structural components in the cortical microtubules. not only plays a role in cytoskeleton support, but also in the movement of cells, form and maintenance of cytoplasmic. Developed adoral ciliatures and cytoskeletal microtubules in the ciliated cells provides a relevant basis for classification in the ciliate svstem.
