The growth status of chlorella and the difference of cell morphology induced by different inoculum densities was investigated in this research. In the experiment, the growth status of chlorella was measured. Meanwhile, the fatty acids content of chlorella as the function of the different inoculum densities were also tested. Based on the GC/MS, the analytic method of high throughput and sensitivity extracting method was established through GC/MS.The dominant research results are: (1) the influence of the growth rate of chlorella induced by different inoculum densities was measured. In comparison with the besides three determination methods was adopted to conduct the growth status of chlorella, which benefits the future study. (2) Through the orthogonal experiment, the method of rapid detect the fatty acid content of chlorella by nile red was optimized, and the optimal condition was using acetone as the solvent, incubated at 25℃10min in the dark,excited at 550nm, emited at 605nm. (3) The analysis of the fatty acid content by GC/MS was conducted, noting that the fatty acid of chlorella which is polyunsaturated and the total fatty acid per milligram dry cellwas changed along with inoculum densities. (4) The high throughput and sensitivity extracting method of metabolites based on the GC/MS was established. By compared with this method, several facters should be taken into consideration before achieving the proper method. Moreover, all metabolites produced by chlorella cannot be extracted by the same method.
