Protein phosphorylation is one of the most important signal transduction pathways in organism. Most function of the phosphatase and the ralationship between protein phosphorylation and signal transduction pathways are not clear because of the superposition between genes. PP2Cs are a member of protein phosphatases family, and it is different from other protein phosphatases in biochemistry quality and the structure. However, it plays an important part in sinal transduction pathways, and in ABA signal transduction pathways which plays a key role in plant responses to abiotic stress. ABA regulates many agronomically important aspects of plant development, including the synthesis of seed storage proteins and lipids, the promotion of seed desiccation tolerance and dormancy, and the inhibition of the phase transitions from embryonic to germin ative growth and from vegetative to reproductive growth. We have read many literatures about PP2C gene family, and found that there were not more studies about PP2C gene family. We acquired PP2CA2 T-DNA homozygous mutant by screened and identified them through the "double-primer method" and the RT-PCR. According to the results of gene chip on Arabidopsis.org website, we studied changes of PP2CA2 gene in wild-type after ABA and Nacl stress treatment.The result shows that, the gene transcription level of PP2CA2 by the induction of ABA and Nacl. We also studied the effect on other stress-related gene expression in ABA signal pathway caused by the deletion of PP2CA2 gene. So we initially identified the probably the location of PP2CA2 gene in the ABA signaling pathway. We observe the changes in phenotype such as seed germination rate, root length, stomatal aperture between the mutant and wild-type. Follow-up test of the physiological studies have shown that mutants are less sensitive to exogenous ABA and Nacl in seed germination and root elongation compared with wild-type. After different concentrations of ABA (0, 0.1, 0.3, 0.6 uM) treated, we discovered that when the ABA concentration is higher than 0.3uM, the mutant seed germination rate was still maintained at the scope of 60%-80%. Further evidence showed that the role of PP2CA2 gene response to salt and ABA signaling pathways in Arabidopsis, which proved that its activation role as a stress-related transcription factor.In order to further explore the function of PP2CA2 gene, we try to construct a sub-PP2CA2 gene over-expression transgenic plant, analysis was proceed on the seed germination rate, root length and stomatal aperture with the over-expression mutants.This work provided some invaluable references for the future research work.
