PurposeThe present study was designed to pursue the kinetic characteristics of acrosome reaction(AR) of sperm in the fallopian tube in vivo, to evaluate the effect of human oviductal epithelial cell(LIOEC) on the AR capacity of sperm and the possible mechanisms through the application of coculture system with HOEC in vitro.The second purpose of this study was to assess the influence of follicular fluid(FF) on the AR capacity of sperm through observing AR rate of sperm preincubated for different time to get capacitation.Methods1.The sperm non-preincubated and the sperm preineubated for different time were incubated in the Ham F10 medium with 20% FF. The AR rate were assesed at 0, 3, 6, 9 hours.2.The sperm were co-cultured with L[OEC and co-incubated with conditioned medium of HOEC. The AR rate were assessed at 0,3,6,9,24 hours. The viability rate of sperm were assessed at 0,3,6,9 hours.3.The sperm were incubated in the medium with 20% FF for 3 hours after cocultured with LIOEC for 3 and 6 hours. The AR rate were assessed.There were comparison groups and control groups in all experiments.Results1.After 3 hours incnabation in Ham F10 medium with 20% FF, the AR rate of sperm non-preincubated,non-capacitated was 5.46% ?.8%, the AR rate of sperm cultured for 3 hours to get capacitation was 20.44%?.9%.2.The spontaneous AR rate of sperm from semen qualified the WHO normal semen criterion and cocultnred with HOEC increased from 3.52% ?.1% at Ohr to 13.05% ?2.3% at 24 hour. But from 6hr after the AR rate were significantly lower than the control (P41.05) . From 3hr after the viability rate of sperm cocultured with HOEC were significantly higher than the control (P41.05)3.FF significantly induce the sperm cocultured with tIOEC for 3 hours and 6 hours3and washed to remove the coculture medium to undergO induced AR(P<00()1).After contacted and reacted with FF the AR rate increased 28.21% and 27.48%respectiveIy.In case the 8perm had not been washcd befOre cont8ctCd with FF theAR rat6 only increa8e l0.57% and l0.68% respectively and were significantlylower (P<0.00l).4. Two major different kinetic patterns have been observed with 8perm of lowquality(not qualify the WRO'8 normal semen criteriOn) 8emen cocultured withHOEC.One major 8ubgroup was so called quickreaction,which was charaterizedby quick spontaneous AR.They underwent the AR immediately after removing thesenimal plasma and attained the maximum value 29.56%l7.2% at 6kr .Anothermajor subgroup was 8o called slow-reaction and Iow AR rate, the maximaI ARrat6 was l0% Iower.5.Two different AR pattern8 had been observed with sperm of low quality semenwith slow AR and Iow AR rate when react6d by FE (l)the induced AR rate ofsperm pre-cocultured with HOEC fOr 3 hours and 6 hours were 21.l0% and26.25% respectively and increased l7.73% and 20.93% than that of 8perm notcontacted with FEthere was 8ignificant difference compared with control(P<000l ). (2)The induced AR by FF were lower than l0%.Conclusions'1.FF migbt have no direct effect on the non- p reinc u bate d, n on-ca pac ita ted s p e rm toundergO AR but it certainly induced sperm preincuted to undergO AR in 3 hours.2.ROEC might prolong 8perm survival, enhauce capacitation and maintain theacro8omal integrity3.Sperm frOm different quality 8emen have different AR kinetic pattern.4.ROEC might enhance the functional perfOrmances of AR of 8perm from lowquality 8emen.
