Objective FMMU albino guinea pig is a new breed of guinea pig. The previous studies suggested that the auditory threshold of FMMU albino guinea pigs was lower significantly than that of pigment ones,the anoxia tolerance of FMMU albino guinea pigs was better than that of pigment ones,their immunological function was weaker than that of pigment ones. The purpose of this experiment is studying RFLP of mitochondrial DNA (mtDNA) from FMMU albino guinea pigs and comparing it with that in pigment ones to find whether it's biological characters are correlative to mtDNA.Methods1. The mtDNAs from FMMU albino and pigment guinea pigs were extracted with alkaline lysis.2. Purified mtDNAs from FMMU albino and pigment guinea pigs were analyzed with 12 restriction endonucleases (Ava I,Bal I,BamH I,Bgl I,Cla I,Dra I,EcoRV,Hpa I,Pst I,PvuII,Stu I,Xho I) and restriction fragment length polymorphism (RFLP).3. The length of mtDNAs and the restriction fragments was calculated with gel image analysis system.Results1. The length of mtDNAs from FMMU albino guinea pigs was the same as that from pigment ones,about 16.7kb.2. The mtDNAs from FMMU albino and pigment guinea pigs had the perfectly same restriction maps,but no polymorphism. The number of recognition site of 12 restriction endonucleases on these mtDNAs was from 1Conclusion1. The length of mtDNAs from FMMU albino guinea pigs was the same as pigment ones about 16.7kb.2. The mtDNAs from FMMU albino and pigment guinea pigs had no polymorphism.3. Special biological characters in FMMU albino guinea pig weren't correlative to mtDNA.
