Rhododendron L. colonized in the North Temperate Zone flora is one of important groups and world famous mountain wild flowers and plants, possessing the very high value to see and enjoy, and the economic value of the medicine.There are 900 much more primitive Rhododendron L. species in the word. For so many Rhododendron L. species, the high probability of interspecific natural hybridization and the high variation frequencies of species and genus groups, researching diversity of Rhododendron L. species must be based on DNA level research.RAPD molecule marker technology was carried on preliminary research to taxonomy of 11 Rhododendron L. species, and its result was identical with morphologic taxonomy basically. Thus it laid a solid base on further study next step for evaluation and taxonomy of new species and variety. 2 × CTAB improved method was used to extract the total genome DNA of high quality of 11 Rhododendron.L species; Rhododendron.L species RAPD optimized reaction system was set up with R.jinggangshanicum Tam, and could be used in 11 Rhododendron.L species RAPD analyses through examining; Moreover 71 kinds of primers (from S10 to S80) were used from Shanghai Sangon, thus 18 kinds of amplified polymorphism primers were gotten. Finally, UPGMA method was used to carry on cluster analysis by NEI-LI formula and Probiosysl.O software.At the same time, 8 Rhododendron L. species' total genome DNA of high quality in which RNA was gotten rid of ,with improved 2× CTAB method, were extracted; through reacting by PCR, about 700bp were amplified, and PCR products were sequenced by ABI310 sequencing machine and these sequences were corrected by man; Together with 11 Rhododendron L. species downloaded from GenBank, nrDNA ITS sequences of 19 Rhododendron L. species were analyzed by PAUP 4.0b4a.The paper pointed out that taxonomy of section of Rhododendron Subgenus Hymenanthes and Subgenus Azaleastrum was accorded with classic taxonomy. But taxonomy of subsection in section didn't be separated, which was related with the poverty of informative site of ITS.
