In order to resolve the sources of preantral follicular oocytes, two methods of isolation were developed in our experiment., The mechanical method resulted in 12.6+3.6 preantral follicles per ovary, The average number of preantral follicles derived from mechanical method was significant lower than that from enzyme associated mechanical method. (p<0.05).This experiment choice the best basic culture system was DMEM+5.00mg/L Insulin+5.00mg/L Transherrin+5.00mg/L Seleniom+2.00mg/L cAMP+1.50mmol/L Glutamone+2.00mmol/L polylysosine+2.00mmol/L Hypoxanthin+5%SerumoExperiment compare to the influence of different culture methods on the preantral follicle cultured in vitro. The result indicates: after 6days cultured, the increments of oocyte in diameter were 29.8 and 32.0μm ,there were no significant differences (p>0.05),but the survival rate from polylysosine layer method no significantly differences than that from ordinary culture method(p<0.05).Experiment add to the different dose EGF and IGF—1 to the basic culture system from to the to cultured the cattle preantral follicles in vitro, the result indicates: EGF and IGF—1 can promote growth and survive of the preantral follicles.Experiment add to the different dose FSH and 17β-E2 to the basic culture system to culture the cattle preantral follicles in vitro , the result indicates: FSH and 17β-E2 can promote the preantral follicle growth and survival. When add to 0. 25mg/L FSH and 1.00mg/L, the result is the best .Experiment search the development after the preantral follicles were cultured for 12 days, the results indicate: after 12 days culture, the mean increments of oocyte in diameter were 62.84μm, there into , 18.76% can form the antrum.
