In recent years, demands for berberine are growing with development on pharmacology study of beberine. but we can't get enough berberine provision because we only depend on two breeding plant, which are coptis and Phellodendrom. To satisfy the demands of markets for berberine, it is necessary to explore the other berberine resource .Mahonia is Chinese medicine which contains berberine, which mainly distributes in the south-west china. Many species distribute in Sichuan .so to study Mahonia in Sichuan is helpful to solve the problems of berberine provisions in Sichuan, even the whole country.We have broadly collect Mahonia plants which distribute in Sichuan province, studied the contents of alkaloids in stem, and studied the tissure culture of Mahonia. the main experimental results are as following::1, during the course of collecting wild plants, 2 species were found recently ,which enrich the records of species of Mahonia in Sichuan province .by now there were 30 species recorded in Sichuan province in total, as farther proves that Sichuan province is distributional centre of Mahonia plant in china, in addition, author found out that wild Mahonia plant resource has been destroying seriously, so it was necessary to take some measures to protect wild resource.2, HLPC was applied to determination of the three kinds of alkaloids including berberine, jatrorrhizine, palmatine in Mahonia stem from different areas in Sichuan province .results showed that (1) berberine, jatrorrhizine and palmatine all exist in Mahonia plant in Sichuan province, berberine was playing a leading role in the compouds of the alkaloid of Mahonia in Sichuan province, jatrorrhizine was second ,palmatine was third; (2) the alkaloid contents of Mahonia in Sichuan were generally high, the totalcontents of three alkaloids was between 1.322% and 2.859%, average 1.833%; the content of jatrorrhizine was between 0.333% and 1.2664%, average 0.725%; the content of palmatine was between 0.099% and 0.725%, average 0.327%; the content of berberine was between 0.289% and 1.584% , average 0.781%; (3)for all samples, coefficient of variation (CV) of each alkaloid was big, first was berberine, second was jatrorrhizine, third was palmatine; but CV of three alkaloid was relatively little ,which was 0.234; (4) difference of three alkaloids total contents of the same specie wasn't obvious according to habitats, but given compound fluctuated greatly, first was berberine, second was jatrorrhizine ,third was palmatine. The research results will conduce to the exploration of Mahonia plants.3^ In the experiment, we induced callus with the M.eurybracteata. the results showed (l)it is easy to form callus with M.eurybracteata explants, under given hormone level, callus can been induced successfully on both MS and B5, which were used as base medium; (2) with induced effects, B5 was obviously more excellent than MS, average induced rate of B5 was 44%, while average induced rate of MS was only 37.4%; (3)when 2, 4-D was 1.0 mg /L and KT was 0.2mg /L ,both MS and B5 showed best induced rate, which induced rate of MS can reach 49%, and that of B5 can do 60%; so the optimum medium was B5+2, 4-D 1.0 mg /L+ KT 0.2mg /L; (4) during whole course of culture phenomena of browning was serious, average browning rate of MS can reach 42%, and that of B5 can do 44%; but high browning rate had no obvious relationship with base medium and hormone level; (5) under optimum condition that the base medium was B5, plus 2, 4-D 1.0 mg /L, KT 0.2mg /L, we contrast induced effects between different explants, results show the tender stem was better than tender leaves. The research results can serve as the base of Mahonia exploration on biological technology.
