Bacteriorhodopsin (BR), a biomolecular material, has both photochromic and unique photoanisotropic properties. This photosensitive protein holds the all-around advantages compared with other organic photochromic materials. Recently, the incorporation of genetic technology infuses new vigor into the application research based on bacteriorhodopsin. Focusing on the holographic properties and applications of bacteriorhodopsin film, the author mainly completed the following works:1. The polarization holographic recording properties of BR were theoretically analyzed. The method of Jones Matrix was used to analyze the diffraction characteristics of holograms taken under different polarization combinations of the object and reference beams. The polarization state and diffraction efficiency of the diffractive beams for different types of polarization holograms were mathematically derived. The dependence of diffraction efficiency kinetic curves on the auxiliary purple light intensity and the readout beam intensity for parallel polarization recordings were theoretically analyzed and computed.2. Using polarization holographic four-wave mixing optical setup, the diffraction efficiency kinetic curves for different types of polarization holograms recorded in BR-D96N film were respectively measured. The transmittance changes of the light intensity passing through the BR-D96N film originated from photochromism and photoanisotropy were also respectively measured. The maximal diffraction efficiencies for parallel polarization hologram and orthogonal polarization hologram were theoretically calculated and compared with the experimental results. The influence of the object-to-reference beams intensity ratio, the intensity and polarization state of the auxiliary purple light as well as the object-to-reference beams intensity ratio under existence of the auxiliary purple light on the diffraction efficiencies were experimentally investigated.3. Using 90-degree geometry of angle multiplexing scheme, six holograms were recorded in the BR-D96N film and the reconstructed images were respectively readout clearly without crosstalk. Based on the polarization holographic properties of BR,polarization multiplexing scheme was implemented in the BR-D96N film. Two images with orthogonal polarizations were recorded as two different holograms; which could be selectively retrieved.4. Using Fourier-transform holographic recording method; the orthogonally circular polarization holographic binary data storage was made on 60 (am x 42 urn area of the BR-D96N film. High signal-to-noise ratio of reconstructed images was obtained The binary data could be decoded without errors. The areal storage density reached to 2 X108 bits/cm2 .5. Preliminary study on holographic interferometry by using BR films was carried out. The interferogram properties obtained by double exposure interferometry and single exposure interferometry were theoretically analyzed. Preliminary experimental results for double exposure and single exposure interferometry on BR-D96N film were presented.
