With the development of fluorescent dye synthesize technique, the bio-label method based on the fluorescent dye is gradually replacing the isotope label method and has been widely used in the field of biotechnology. Because of the high molar extinction coefficient and quantum yield, the cyanine dyes as a new fluorescence probe have gotten important application in the research fields of life science, such as microarray-based analysis, DNA sequence, fluorescence immunoassay, flow cytometery, clinical diagnose and so on. In this thesis, the mechanism of the preparation of electronegative fluorescent dye doped core-shell fluorescent nanoparticles has been studied under the combination of the advantages of cyanine dyes and preparation technique of core-shell fluorescent nanoparticles. Cyanine dyes doped fluorescent nanoparticles have been prepared based on this mechanism. A fluorescence label method has also been developed by use of the cyanine dyes doped fluorescent nanoparticles, which provided a new idea for the application of cyanine dyes doped fluorescent nanoparticles in biomedicine. Study on the preparation of electronegative fluorescent dye doped core-shell fluorescent nanoparticlesThe effects of different protein or peptide modified Cy3, Cy5 or FITC as the core material for preparation of fluorescent nanoparticles with stable core-shell structure have been investigated by use of the electronegative organic fluorescent dye Cy3, Cy5 and FITC as demonstration. The results indicated that very bright and photostable Cy3, Cy5 or FITC doped core-shell silica fluorescent nanoparticles can be prepared respectively by use of poly-lysine or IgG modified electronegative organic fluorescent dye as the core material because the isoelectric point of the two kinds of biomolecules is both over 7 at neutral pH. And the dye leakage from the nanoparticles is also very little. The results not only offered us theory foundation for preparation of electronegative organic fluorescent dye doped silica fluorescent nanoparticles but also open up a new idea for developing the application of cyanine dyes in the field of biology technology.Partâ…¡.
