In order to study the biology characteristic of human placenta derived mesenchymal stem cells and its functions in the cord blood hematopoietic stem cells transplantation, we used enzyme digestion and density gradient centrifuge to separate mesenchymal stem cells from the mature placenta, and appraised its general biological characteristic and multi-differentiation potential. We detected the expressions of hematopoietic factors in MSC with RT-PCR. In this foundation, we established in vitro expansion system of cord blood CD34~+ cells which is based on human placenta derived mesenchymal stem cells, studied the effects of MSC on the expansion of cord blood CD34~+ cells, and discussed its initial molecular mechanisms. The results indicated that, the MSCs derived fromhuman placenta were homogeneous and have multi-differentiation potential which may differentiate to fat, bone and cartilage cells. They expressed many kinds of hematopoietic factors, including SCF, FL, G-CSF, GM-CSF, M-CSF and IL-6. Obviously, they increased the expansion of cord bloog CD34~+ cells in vitro. The results indicated that, MSCs which were separated from human placenta could be used for tissue engineering, gene therapy and cell treatment. Placenta MSCs could be used for the matrix of cord blood hematopoietic stem cells, which were suitable for the microenviroment of cord blood hematopoietic stem cells. One the basis of our findings, It is hopeful in the process of implant to reconstruct the hematopoietic microenvironment, promote expansion of hematopoietic cells, shorten the time of hematopoietic comeback, and reduce GVHD, and we may transplant cord blood with placenta MSC, which enable cord blood transplant clinically to have more widespread application prospect.
