The development of bait algae culturing and laser technology used in this field were presented in this paper. The effective method and experiment parameter of laser irradiating Platymonas Subcoydiformis was researched, using various kinds of laser irradiating Platymonas Subcoydiformis. In the same time, the biological effect of Platymonas Subcoydiformis induced by laser irradiation was detailedly studied in virtue of absorption spectrum, Raman spectrum and Laser Scanning Confocal Microscope, LSCM. The research result provides an experiment foundation for the field of laser employing in alga research and experiment base for laser biological effect mechanism research.Firstly, the status of Laser technology used in algae field was introduced briefly. The difficulty of algae culturing and laser biological effect were pointed out and then the paper's research method was given.Secondly, the influence induced by laser's parameter, including dose, wavelength, power and polarity, during laser irradiating Platymonas Subcoydiformis, was studied by the numbers. The experiment result show that there were the best dose ,the most functional wavelength and the best combination of irradiating time and power in the course of laser irradiating Platymonas Subcoydiformis; Polarization or non-polarization has no different result.Thirdly, the auto fluorescence image and spectrum of Platymonas Subcoydiformis were gained by LSCM. A further research was developed between LD laser, Nd:YAP laser irradiating algae and the control group, in order to find the different in the auto fluorescence image and spectrum.The result show that laser irradiation expand the area of chloroplast, enhance its auto fluorescence intensity and activate photosynthesis. Low-intensity laser irradiation can improve the activity of collagen protein, but high-intensity functioned conversely.Finally, Raman spectrum and infrared absorption spectrum of Platymonas subcordiformis were studied by FT-Raman spectroscopy and infrared absorption spectroscopy. The precise measurements of algae Raman Spectroscopy could be used for research of biological samples, such as developing a new optical taxonomic methodology to distinguish different algae species, and a rapid, non-destructive detection way of stress effects.
