Arabidopsis thaliana is the most important material and plays an important role in the study of plant development and molecular biology. It is meaningful to study the establishment of regeneration system and cell suspension system of Arabidopsis thaliana.The effect of environmental stresses on the growth and development of plant cells and tissues becomes a hot research field. As one kind of environmental stresses, ultrasound has very important effect on the plant growth and development: low intensity ultrasound can accelerate the growth and affect the growth of plant or some organs as well. We treated the cell suspension system by the ultrasound instrument to study the biologic effect resulted from the ultrasound on Arabidopsis thaliana by studying the physiology and biochemistry change, which provide the theory support for ultrasound biologic effect.1. A regeneration system was established. We chose the stem of Arabidopsis thaliana as the original material for explant. By adding different kinds and levels of hormones, the materials were induced into forming callus, bud and root, we found that B5+1mg/L 6-BA+0.3mg/L NAA, MS+5mg/L6-BA+0.5mg/LNAA, and l/2MS+lmg/L 6-BA+0.5mg/LNAA+0.1mg/L KT were respectively adapt to induce callus, bud and root.2. The yellow, loose, fresh callus was chosen for cell suspension culture. The component of culture medium was B5 basic culture medium, adding 0.5mg/L 2,4-D+0.05mg/L 6-BA, sucrose 20g/L, pH5.5. After cultivated for one month we got an eugenic suspension cell culture system.3. During the growing of cell suspension of Arabidopsis thaliana, the pH, sugar concentration and activity of SOD and CAT were tested by measured the cultivated liquid every 2 days. Results showed that the growth style of cell suspension of Arabidopsis thaliana presented S-type. The consuming of sugar and the down of pH was obvious. The growth of cell was bloomed, inducing high SOD activity and the peak activity of CAT on the 11th day.4. When the cell suspension of Arabidopsis thaliana was treated by ultrasound, treatment in 1 minute accelerated the growth of cells temporarily. We...
