Screening And Cultivation Of The Streptomyces Species Of Highly Productive MV5002 And Preliminary Study On The Melanin

The melanin is a sort of pigments which wildly exists in propagation and microorganism.Nicolaus et al suggested melanins that can be classified into three clusters according to the chemical constitution: eumelanins,phaeomelanins and allomelanins.For its extensive application, the researche about the microorganism produced melanin and its application has got increasing concern. In the latest years, researchs on producing melanin through microorganism ferment increases continuously in China. Our laboratory obtained a melanin producing MV5002 by gathering and isolating soil from different areas.Having observed MV5002 morphologically comparising and characteristization, analyzing the component of cytochemistry and the physiological and biochemical reaction, were identified this bacterium potentially belong to strain of the Streptomyces antibioticus .To improve the melanin′s product of the MV5002, the fermention condition and medium of the MV5002 were optimized.The result indicates that the most suitable medium is : glycerol 1,beef extract 10,NaCl 6,CaCl2 0.05,L-tyr 1.5,(g/L)pH 8.0, At 31℃and 280 r/m,the output of melanin reaches 3.45 g/L which is 2.88 times of the output under the primary ferment condition.Preliminary study on the melanin′s physical-chemical...
