Sleep disorders can effect the human's life and working. long-term sleep deprivation can effect the functions of development and Immunity .at the same time, it can damage the ability of learning and memory. lots of scholars have been study the relationship deeply between sleep and learning and memory, meanwhile they provid many experiment evidences. up to now, the real mechanism about sleep disorders leads to learning and memory lesion does not discover completely.In order to know the function's mechanism of sleep disorders lead to the space learning and memory descent,we designed the following experiment to study it,moreover,we study the role of intervention to sleep disorders by Oryzanol and multi-vitaminB.The first part: rat model of sleep deprivation was established by intracerebroventricular injection of PCPA and atropine. The electroencephalogram (EEG), electromyogram (EMG), eletrooculogram (EOG) were recorded and the data will be disposed automodicly every 30 seconds by analysis programme which was writed by the visual basic language. In every 24 hour, the total sleep time, NREM time and the REM time of control group is longed to the sleep deprivation group, but the waking time of control group is shoted to sleep deprivation group. Above results indict that the rat sleep deprivation model was established . The second part: Utilizing the Morris water maze test the ability of rat's space learning and memory; puting the technology of brain slice patch clamp whole-cell recording to study the long-term potentiation (LTP) of CA1 region pyramid cells, simultaneous, using the technique of intercell marking to ascertain the cell's form. The main results are as follows:1. In the Morris water maze test, escape latency to find a hidden platform increased and the times of crossing target platform and the percent of swimming in northeast obviously decreased in sleep deprivation group. It indicates the control group's performance of spacial learning and memory was better to the sleep deprivation group. 2.The NMDA receptor-mediated LTP of sleep deprivation group was inhibited, there is no difference...
