The irradiation of ultraviolet can induce oxidized-lesions like lethal pyrimidine dimers in organism genomes. One significant lesion is the 6-4 pyrimidine pyrimidone photoproduct(6-4 PP). It directly causes apoptosis, thus a remarkable inducer of mutation and cell death. On the other hand, cyclobutane pyrimidine dimmer(CPD) just has an effect of cell cycle arrest. Therefore, the research and utilization of the 6-4 photolyase gene is of predominant significance.Dunaliella salina is a salt-tolerant unicellular green alga which can survive extreme salty environment and possesses comparative tolerance against UV.We had reported that the 6-4 photolyase gene Ds64PHR of Dunaliella salina had been successfully cloned for the first time. On that groundwork, we did a series of research on this gene focusing on properties of lesion-repair performed by the gene product 6-4 photolyase and gene expression at the level of nucleic acid and protein.1 , The function of the gene Ds64PHR was identified in vivo through complementary experiment.
