After cytochrome C extracted from Qinchuan cattle heart muscle by acid liquid water,Permutet absorption,ammonium sulfate salting out,TCA precipitating,dialysis and filter,its content was measured further.The cytochrome C was purified by D-85 ion-exchange resin in chromatography column.Iron content and enzyme activity of the cytochrome C were determined.Purity was tested by electrophoresis too.After extracted cytochrome C form cattle heart muscle,coenzyme Q10 was extracted by alcohol base saponification,petroleum ether extracting,condensing,silica gel colume,ethyl alcohol.Coenzyme Q10 was distinguished by thin-layer chromatography in comparison with Rf data.Coenzyme Q10 purity was determined with ultraviolet spectrophotometer at 275nm.The samples of extract purity were analyzed.The important results of the study showed:(1) The technology of extract purity cytochrome C was reasonable.The sample was one spectrum band by electrophoresis.The sample and the standard were scaned and absorption spectrum were the same.There were max-absorb-peak in 520nm and 550nm.(2) The cytochrome C were determined and average retrieve rate was 146.3mg/kg.The result was higher than internal report's.The iron content of the sample of purity was average 0.42% and enzyme activity was average 96.2%.The results of analysis conformed to china medicing model.(3) Coenzyme Q10 was prepared form cattle heart muscle surplus.Coenzyme Q10 saponification suitable temperature was 90℃,and soponifieation time was 30min,and abstract number of times was two.Coenzyme Q10 was only one stain by thin-layer chromatography,Rf=0.56(a=7.5cm,C=13.5cm).The sample and the standard were scaned and absorption spectrum were the same.There was max-absorb-peak at 275nm.The sample purity was 91.2%.(4) Based on cattle heart muscle physiological characteristic,adopting pH3.4 acid liquid water to extract the cytochromec,using D-85 large-pore ion-exchange resin to substitute for Amberlite IRC-50 ion-exchange resin,the resalt showed that effect of purity was better.(5) Adopting items biochemistry technology,the cytochrome C was first extracted from cattle heart muscle,and then coenzyme Q10 was extracted from surplus materials,which will offer technology and the method to syntheticly apply resource in accord with the demands at present saving society.
