The genus Microcystis is known to be a toxic and water bloom-forming Cyanobacterium.Microcystins are a kind of endotoxins, which are strong carcinogens. Drinking water-containedMicrocystins for a long time may cause liver damages on human beings or animals.Beauty spots play an inportant part in people' s life from winnter to autumn.However, Microcystiscan destory zoology sight and tourism of the beauty spots.What' s worse, Microcystis can produceMicrocystins,which is a tumor-promoting agent.The toxin,not only destory the ecologicalstructure,but also endanger people's life,has been blocked up the development of communityeconomy and environment.Common detection measures, such as HPLC and ELISA, can detect Microcystins in water. But theycan not monitor the producing of Microcystins before they are released from the cell of Microcystis.Based on 16S rDNA sequences of Microcystis, Cyanobacteria and Microcystin synthetase gene B(mcyB), primers for amplification and sequencing were designed.A duplex PCR and nested PCRwere used for the detection to study the seasonal distribution of toxic Microcystis from eight beautyspots, including 108 water samples in Guangzhou of Guangdong province in three seasons. Thebeauty spots were Liuhua park,Yuexiu park,Baiyun mountain,Dongshan park,Xiaogang park,Tianhe park,Liwan park and Jinan University.Results showed that total positive ratio of toxicMicrocystis was 38.9% in winter, 94.4% in summer, while 92.5% in autumn. The data showed thattoxic Microcystis distribut in winter, summer and autumn. Microcystins pollutions in Guangzhoubeauty spots was mainly in summer and autumn, because the temperature illumination and thepressure of population competition were fit for the growth of Microcystis, but which be not inincluded in winnter.
