A medicinal plant Huperzia serrata (Thunb.)Trev. has been selected to isolate its endophytic fungus, screened the bioactive strains and studied the strain with better bioactivities. The results were as follows:1. There were 180 strains of endophytic fungus isolated and purified from the roots, stems and leaves of Huperzia serrata by the technique of tissue isolation. They were identified as 2 subdivsions, 4 classes, 7 orders, 8 families, and 13 genus. Phoma sp., Phacodium sp. were dominate genus in Huperzia serrata, their percent were 18.61% and 16.15% of the all strains. It showed that the quantities, species and distribution of endophytis fungus varied with different parts of Huperzia serrata. The most of them were in stems and the least were in roots. The number of endophytic fungus separated between 4th-5th day and 11st-13rd were comparatively more.2. The bioactive strains were selected by screening their bioactivities on five models, such as hydrolyzing fibrin, bacteria, pathogens, cancer cell and HPLC. And 68 representative strains were chosen from the all.3. Because of its better bioactivity, we studied the ITS of HU046 and the results showed that it belongs to Phoma sp..4. The optimal culture conditions for the growth of HU046 strain were as follows: the optimal temperature was 26℃, the optimal pH was 4.0, the optimal illumination was dark, the optimal carbon and nitrogen source were corn flour and yeast ointment and the optimal C/N was 30:1.5. The volatile oil from the broth of HU046 was analyzed by GC-MS, 11 compounds were identified from it and the results showed that its main components were alkyls,ketone,phenols and esters.6. Metabolites of the HU046 were isolated by the isolation technique for natural products. 3 compounds were obtained from the second metabolites of strain HU046, and 1 compound was elucidated as mannitan by nuclear magnetic resonance technique.7. The HUO46 was analyzed by TLC and HPLC. And the results showed that the strain tested might have the component huperzine A or the analogue of huperzine A .
