Forkhead box (Fox) proteins play critical roles in the regulation of differentiation,proliferation, immunity and aging of cells. Most studies on Fox proteins are limited tocultured cells and rodent. The aim of the current study is to detect whether FoxO1,FoxO3a, FoxO4 proteins are localized in the stomach and intestine of the pig and ratand changing of expression of FoxO proteins by immunohistrochemistry,immunofluorescence, Western- blot, radioimmunoassay.1. Localization of FoxO proteins in pig's gastrointestinal tracts.Four adult commercial pigs of Duroc(Landrace(Large Yorkshire)) three-waycross combination were used in the current experiment. The aim of the study is todetect whether FoxO1, FoxO3a, FoxO4 proteins are localized in the stomach andintestine of the pig by immunohistrochemistry. The results showed that FoxO4 existsin the mucosa in all parts of the stomach and intestine. FoxO3a exists mainly in thelamina propria and muscularis of some parts. However, FoxO1 is not detectable in allparts of the stomach and intestine. Collectively, the results of the present studyindicate that there exists a distinct expression pattern of Fox proteins, and that FoxO4is a primary forkhead transcriptional factor localized in the gastrointestinal tracts ofadult pig.2. Aged- dependent localization of FoxO4 in pig's gastrointestinal tracts.As the previous studies, FoxO4 is the main forkhead transcriptional factor ingastrointestinal tracts of adult pigs. In order to detect changing of FoxO4 expressionwith the development in gastrointestinal tracts, at least, three pigs in each four agegroups of Landrace-Duroc F1 cross breed commercial pigs including 0d, 30d, 160dand 180d were used. The study detected aged- dependent localization of FoxO4 by immunohistrochemistry.3. Aged- dependent expression of FoxO4 in rat's gastrointestinal tracts.The purpose of this study was to validate whether FoxO4 expressed ingastrointestinal tracts of rats, rodent animals, was aged- depended which similar inpigs. Experiments were conducted to examine aged-dependent expression of FoxO4proteins in the duodenum of 21d, 2 months and 6 months rats byimmunohistochemistry and Western- blot. The results indicate that expression ofFoxO4 in rats' duodenum is aged- depended, localization specialization and tissuespecialization. The work researched changes of FoxO4 protein in development whichprovided prerequisite for FoxO4 protein functions researches in gastrointestinal tractsGenerally, FoxO4 is the main forkhead transcriptional factor in adult pigs'gastrointestinal tracts; as the downstream of PI3K/AKT pathway, FoxO proteinsexpress differently in pig and rat's gastrointestinal tracts which have closelyconnection with animal development and metabolism.
