Partâ… The human ZNF313 gene is a novel spermatogenesis-related zinc finger protein gene that contains both C2H2 and C3HC4 domains. It is particularly important to study the expression and function of this gene and its homologue gene.We have cloned a novel mouse zinc finger protein gene Znf313 by rapid amplification of cDNA ends (RACE) according to the homologue of human ZNF313 gene which contains a C3HC4 ring finger domain and three C2H2 domains. Revealed by Northern blot, the full mRNA consists of 2.16 kb and is widely expressed in tissues as a single transcript. For better understanding the expression of mouse Znf313 gene, we analyze its expression in many tissues even in early embryo by RT-PCR. Western blot and subeellular localization protocol were also used for inspecting the space of its expression product. Revealed by RT-PCR, the expected 364 bp PCR product of the Znf313 gene was ubiquitously expressed in all tissues and the expression of Znf313 in testis is detected in all development stages. Western blot analysis also reveals that Znf313 is expressed in the tissues, subcellular localization demonstrate that Znf313 is expressed both in the cytoplasm and nucleus whereas predominantly localized in the nucleus in both HEK293T and 3T3 cells. Present data suggests that Znf313 gene might play a fundamental role in gene transcription and regulation in organism and related to spermatogenesis.Partâ…¡It is conservation for biology during the process of evolution. Research has revealed that the homologue genes always have the same or similar function, so it makes the mode animal become an important plat for studying the function of genes.We have cloned a novel zebrafish zinc finger protein gene Znf313 by rapid amplification of cDNA ends (RACE) according to the homologue of human ZNF313 gene. The cDNA is 1,241 base pairs (bp) in length and encodes a 221 amino acids (aa) protein with a C3HC4 ring finger domain and two C2H2 domains. 54.56% and 48.91% nucleotide (nt) and aa sequence identity is observed with its human homologue. Revealed by RT-PCR, Znf313 gene is widely expressed in tissues. Using whole mount in situ hybridization with DIG antisense RNA probe, we detected the expression of the zebrafish Znf313 gene during embryogenesis. Cellular localization demonstrates that Znf313 is expressed both in the cytoplasm and nucleus whereas predominantly localized in the nucleus. Present data suggests that Znf313 gene might play an important role during embryogenesis.
