Lactobacillus are Probiotic species, There was not a well perfect method of detection and identification for Lactobacillus yet. This study aims to explore the application of Phenotype taxonomy, 16S rRNA sequences and phylogenetic analysis, and Biolog Automated Microbes Identification System (BAMI System) for 15 strains of three species of Lactobacillus (L. casei, L.paracasei subsp.paracasei, L. rhamnosus) that came from international standard culture collection. The result indicated that Phenotype taxonomy is difficult to identified tested strains to species level; because of high homology of 16S rRNA gene, 16S rRNA sequences and phylogenetic analysis could only identify tested strains to genus level; BAMI System is accurate enough for the identification of tested strains above, but the universality of BAMI System in identification for this three species of Lactobacillus keeps unclear, and more data is needed. The three technique mentioned above all showed respective results in different level of identification. It could provide practicable theory for establishment of quick identification system fit for 3 species of Lactobacillus, used for reference in identification for other species, could be supplied as a necessary qulity control measure for increasingly all-pervading Lactobacillus probiotic products.
