Griffithsin (GRFT) is a novel anti-virus gene which can bind to HIV-1 envelope specially. On this study, the anti-HIV-1 gene GRFT was acquired by the digestion of plasmid pET-GRFT. Recombinant plasmid pGEX-GRFT was constructed by the plasmid pGEX-6p-1 and the gene of GRFT. Then transformed into E.coli BL21 (DE3) strain and induced by IPTG. And the condition of inducing temperature, inducing time and IPTG concentration were screened . At last, the best expression condition was decided. The different target products all existed in the form of inclusion body.The Dot-ELISA was used to detect the activity of the renaturation protein. The renaturated protein takes the reaction with recombinant gp120 antigen. It can be proved the recombinant protein have the biological activity. The conclusion can be the base of researching the resisted HIV praeparatum.
