Osteocalcin (BGP) is one of the few proteins specifically expressed in osteoblast cells, it has several features of the hormone. For example, as a molecule precursor synthesized, it's a specific cell molecules, secreted into the circulatory system. Osteocalcin is rich in bone tissue, it is synthesized and secreted by the mature osteoblast, most of which were deposited in bone matrix and about one-fifth of it released into the blood, the levels of the osteocalcin which in the serum and bone tissue has positively correlated, so the content of the serum osteocalcin in bone tissue can indirectly reflect the synthesis of osteocalcin in the situation. Because osteocalcin occurs mainly in the mineralized formation stage, it is considered to the mineralization of osteoblast differentiation markers of the occurrence period.This paper uses genetic engineering techniques cloning the osteocalcin gene and transferred to the eukaryotic and prokaryotic expression vectors, the experiment through the transformation of pcDNA3.1(+)-HA, pcDNA3.1(+)-HA-His and pGEX-4T-1 plasmid, making the osteocalcin gene labeling with HA, His and GST in the vector, get the following results:â‘ A successful eukaryotic expression vector was built named pcDNA3.1(+)-HA-BGP;â‘¡The eukaryotic expression vector which named pcDNA3.1(+)-HA-His-BGP was successfully constructed with the HA and His tagged;â‘¢The prokaryotic expression vector pGEX-4T-1-BGP was successfully constructed with the GST, it provide an experimental platform and basic materials.for the follow-up experiments and functional protein expression experimentsIt found that osteocalcin also has the function of regulating glucose metabolism in Recent study, in the regulation of glucose metabolism, it can contain fat while enhancing the insulin levels and the absorption of insulin, this new feature could have important impact for diabetes treatment.
