During the development of embryo in vertebrates,pax6 is one of important transcription factors,especially during the process of eye's development.Pax6 is the gene first found both expressed during the process of lens' development and regeneration.Previous studies have shown that pax6 have many different isoforms,and these isoforms control the diversity regulation of pax6.In this paper,we collected the 12-25 stages'embryos of Bufo raddei Strauch,and the 1th,3th,5th,7th,9th,14th,21th,30th,40th days'tissues of lens'regeneration after removed the lens of tadpoles. We exteacted of proteins from the different stages'embryos and different days'tissues of lens regeneration,and determined the concentration of proteins by the method of coomassie brilliant blue G-250,detected the proteins by SDS-PAGE, determined and analysed the expression of pax6 and pax6 variant during the process of lens' development and regeneration by Western blot. The results of Western blot shown that:at the 17 stage of lens development,the expression of pax6 was the highest,the MOD was 0.355; at the 20 stage, the expression of pax6 was distinguished higher, the MOD was 0.229.On the 7th day of the lens'regeneration, the expression of pax6 was significantly higher, the MOD was 0.219;especially on the 30th day,the expression was the highest,the MOD was 0.260. At the 16 stage of the lens development,the expression of pax6 variant was the highest,the MOD was 1.932; at the 22 stage, the expression was distinguished higher, the MOD was 1.486.In the 5th day of the lens' regeneration, the expression of pax6 variant was significantly higher, the MOD was 0.331;especially on the 21th day,the expression was the highest,the MOD was 1.801.The results above shown that:pax6 and pax6 varianr were very important during the process of lens'development and regeneration,and there was a dominant negative effect between pax6 and pax6 variant.In general,the results above laid a solid foundation for further research the precise role of pax6 and pax6 variant in lens development and regeneration, and provided the basis for future studies into the functions of pax6 and pax6 variant.
